Italian Institute Of Health Drastically Reduces Its Official COVID Death Toll Number

The Italian Higher Institute of Health has drastically reduced the country’s official COVID death toll number by over 97 per cent after changing the definition of a fatality to someone who died from COVID rather than with COVID.

Italian newspaper Il Tempo reports that the Institute has revised downward the number of people who have died from COVID rather than with COVID from 130,000 to under 4,000.

“Yes, you read that right. Turns out 97.1% of deaths hitherto attributed to Covid were not due directly to Covid,” writes Toby Young.

Of the of the 130,468 deaths registered as official COVID deaths since the start of the pandemic, only 3,783 are directly attributable to the virus alone.

“All the other Italians who lost their lives had from between one and five pre-existing diseases. Of those aged over 67 who died, 7% had more than three co-morbidities, and 18% at least two,” writes Young.

“According to the Institute, 65.8% of Italians who died after being infected with Covid were ill with arterial hypertension (high blood pressure), 23.5% had dementia, 29.3% had diabetes, and 24.8% atrial fibrillation. Add to that, 17.4% had lung problems, 16.3% had had cancer in the last five years and 15.7% suffered from previous heart failures.”

The Institute’s new definition of a COVID death means that COVID has killed fewer people in Italy than (whisper it) the average bout of seasonal flu.

If a similar change were made by other national governments, the official COVID death toll would be cut by a margin of greater than 90 per cent.

Don’t expect many others to follow suit though, given that governments have invested so much of their authority in hyping the the threat posed by the virus.

For example, behavioral psychologists in the UK worked with the state to deliberately “exaggerate” the threat of COVID via “unethical” and “totalitarian” methods of propaganda in order to terrify the public into mass compliance.

And it worked.

A survey conducted after the first lockdown found that the average Brit thought 100 times more people had died from COVID than the official death toll.

Now we come to understand that the official killed ‘by COVID’ and not ‘with COVID’ figure is less than one tenth what is officially reported as the total COVID death toll.

Despite the change, Italy may yet take the decision to make the COVID-19 vaccine mandatory, although how such a scheme would be imposed remains unspecified.

UK approves Merck’s daily Covid pill… First country in the world to allow use…

(Reuters) – Britain becomes the first country in the world to approve a COVID antiviral pill developed by Merck Britain’s Medicines and Healthcare products Regulatory Agency (MHRA) recommended the drug, molnupiravir, for use in people with mild to moderate COVID-19 and at least one risk factor for developing severe illness, such as obesity, older age diabetes, and heart disease.

It will be administered as soon as possible following a positive COVID-19 test and within five days of the onset of symptoms, the regulator said, citing clinical data.

The green light is the first for an oral antiviral treatment for COVID-19 and the first for a COVID-19 drug that will be administered widely in the community. U.S. advisers will meet on Nov. 30 to review the drug’s safety and efficacy data and vote on whether molnupiravir should be authorized.

The pill, which will be branded as Lagevrio in Britain, is designed to introduce errors into the genetic code of the coronavirus that causes COVID-19 and is taken twice a day for five days.

Drugs in the same class as molnupiravir have been linked to birth defects in animal studies.

Merck, known as MSD outside of the United States and Canada, has said animal testing shows that molnupiravir is safe, but the data have not yet been made public.

“We are compelled to urge you to hold the Committee’s hearing on Prof. Lipstadt’s nomination without further delay,” said the letter from the Jewish Federations of North America, the Orthodox Union and the Anti-Defamation League.

The letter was addressed to the chairman of the committee, Democrat Robert Menendez of New Jersey, and its ranking member, Republican James Risch of Idaho, although its clearly intended target was Risch, who is using his prerogative to hold up a hearing.

Risch told Jewish Insider this week that he was concerned about past Lipstadt tweets in which she was critical of Republican lawmakers. Other Republican lawmakers fault her for appearing in an ad during last year’s presidential campaign likening former President Donald Trump’s rhetoric to those of the Nazis in the 1930s, before the Holocaust.

This week, Lipstadt testified on behalf of the plaintiffs in a civil suit against the organizers of the 2017 white nationalist rally in Charlottesville, Virginia, where Trump famously said there were “very fine people on both sides” and which Biden made a centerpiece of his presidential campaign.

PF-07321332 has been specifically designed to be administered orally, so that it can potentially be administered at the first sign of infection with, or exposure to, SARS-CoV-2, without requiring patients to be hospitalized

NEW YORK(BUSINESS WIRE) Pfizer Inc. (NYSE: PFE) today announced the start of the Phase 2/3 EPIC-PEP (Evaluation of Protease Inhibition for COVID-19 in Post-Exposure Prophylaxis) study to evaluate the investigational novel oral antiviral candidate PF-07321332, co-administered with a low dose of ritonavir, for the prevention of COVID-19 infection. This Phase 2/3 trial is part of a global clinical research program and is enrolling individuals who are at least 18 years old and live in the same household as an individual with a confirmed symptomatic SARS-CoV-2 infection.

“With the continued impact of COVID-19 around the world, we believe that tackling the virus will require effective treatments for people who contract, or have been exposed to, the virus, complementing the impact that vaccines have had in helping quell infections. If successful, we believe this therapy could help stop the virus early – before it has had a chance to replicate extensively – potentially preventing symptomatic disease in those who have been exposed and inhibiting the onset of infection in others,” said Mikael Dolsten, MD, PhD., Chief Scientific Officer and President, Worldwide Research, Development and Medical of Pfizer. “Given the continued emergence and evolution of SARS-CoV-2 variants and their immense impact, we continue to work diligently to develop and study new ways that our investigational oral antiviral candidate could potentially lower the impact of COVID-19, not only on patients’ lives, but also the lives of their families and household members.”

The Phase 2/3 EPIC-PEP trial is a randomized, double-blind, placebo-controlled study and will enroll up to 2,660 healthy adult participants aged 18 and older. Participants will be randomly assigned (1:1:1) to receive PF-07321332/ritonavir or placebo orally twice daily for 5 or 10 days. The primary objective will assess safety and efficacy for the prevention of confirmed SARS-CoV-2 infection and its symptoms through Day 14. PF-07321332 is an oral antiviral SARS-CoV-2-3CL protease inhibitor, which has an encouraging pre-clinical profile, including potent in vitro antiviral SARS-CoV-2 and broad coronavirus activity. Results from the Phase 1 clinical trial demonstrated that PF-07321332 was safe and well tolerated.

In addition to this study, the global EPIC program consists of multiple ongoing clinical trials, including one in SARS-CoV-2 infected patients who are at high risk of severe illness (including hospitalization or death), which began in July 2021, and another in infected patients who are at standard risk (i.e., do not have risk factors for severe illness), which began in August 2021.

Exploring the Binding Mechanism of PF-07321332 SARS-CoV-2 Protease Inhibitor through Molecular Dynamics and Binding Free Energy Simulations

Abstract

The novel coronavirus disease, caused by severe acute respiratory coronavirus 2 (SARS-CoV-2), rapidly spreading around the world, poses a major threat to the global public health. Herein, we demonstrated the binding mechanism of PF-07321332, α-ketoamide, lopinavir, and ritonavir to the coronavirus 3-chymotrypsin-like-protease (3CLpro) by means of docking and molecular dynamic (MD) simulations. The analysis of MD trajectories of 3CLpro with PF-07321332, α-ketoamide, lopinavir, and ritonavir revealed that 3CLpro-PF-07321332 and 3CLpro-α-ketoamide complexes remained stable compared with 3CLpro-ritonavir and 3CLpro-lopinavir. The binding energy of the bespoke antiviral PF-07321332 clinical candidate was two times higher than that of α-ketoamide and three times than that of lopinavir and ritonavir. Our study elucidated in detail the binding mechanism of the potent PF-07321332 to 3CLpro along with the low potency of lopinavir and ritonavir due to weak binding affinity demonstrated by the binding energy data. This study will be helpful for the development and optimization of more specific compounds to combat coronavirus disease.

Abstract

The utility of gabapentin and lamotrigine for the treatment of bipolar disorder is reviewed. Bipolar disorder is characterized by extreme mood fluctuations, including mania, hypomania, depression, and mixed episodes. Extrapolation of postulated mechanisms of anticonvulsant activity in bipolar disorder has led to the use of the newer anticonvulsants gabapentin and lamotrigine for therapy. Both agents appear promising on the basis of limited (often anecdotal) evidence. They may prove effective in patients with difficult cases of bipolar disorder, such as patients with rapid cycling, mixed episodes, and illness refractory to other treatments. Lamotrigine may offer a much-needed treatment alternative for bipolar depression and could be found effective for acute mania, but the need for slow dosage adjustment and the risk of rash may limit overall clinical utility. Gabapentin may offer significant advantages for acute mania: The dosage can be adjusted rapidly, adverse effects are generally minimal, the therapeutic index is high, there is no required laboratory monitoring, and there is minimal potential for interactions with other psychotropics. Until the results of randomized controlled trials are known, however, these two agents should be reserved for patients with bipolar disorder unresponsive to traditional therapies and for patients who cannot tolerate traditional agents. Preliminary evidence indicates that gabapentin and lamotrigine may be useful for the treatment of bipolar disorder.
