Two Different Antibody-Dependent Enhancement (ADE) Risks for SARS-CoV-2 Antibodies

Abstract

COVID-19 (SARS-CoV-2) disease severity and stages varies from asymptomatic, mild flu-like symptoms, moderate, severe, critical, and chronic disease. COVID-19 disease progression include lymphopenia, elevated proinflammatory cytokines and chemokines, accumulation of macrophages and neutrophils in lungs, immune dysregulation, cytokine storms, acute respiratory distress syndrome (ARDS), etc. Development of vaccines to severe acute respiratory syndrome (SARS), Middle East Respiratory Syndrome coronavirus (MERS-CoV), and other coronavirus has been difficult to create due to vaccine induced enhanced disease responses in animal models. Multiple betacoronaviruses including SARS-CoV-2 and SARS-CoV-1 expand cellular tropism by infecting some phagocytic cells (immature macrophages and dendritic cells) via antibody bound Fc receptor uptake of virus. Antibody-dependent enhancement (ADE) may be involved in the clinical observation of increased severity of symptoms associated with early high levels of SARS-CoV-2 antibodies in patients. Infants with multisystem inflammatory syndrome in children (MIS-C) associated with COVID-19 may also have ADE caused by maternally acquired SARS-CoV-2 antibodies bound to mast cells. ADE risks associated with SARS-CoV-2 has implications for COVID-19 and MIS-C treatments, B-cell vaccines, SARS-CoV-2 antibody therapy, and convalescent plasma therapy for patients. SARS-CoV-2 antibodies bound to mast cells may be involved in MIS-C and multisystem inflammatory syndrome in adults (MIS-A) following initial COVID-19 infection. SARS-CoV-2 antibodies bound to Fc receptors on macrophages and mast cells may represent two different mechanisms for ADE in patients. These two different ADE risks have possible implications for SARS-CoV-2 B-cell vaccines for subsets of populations based on age, cross-reactive antibodies, variabilities in antibody levels over time, and pregnancy. These models place increased emphasis on the importance of developing safe SARS-CoV-2 T cell vaccines that are not dependent upon antibodies.

Abstract

The emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants that escape convalescent and vaccine-induced antibody responses has renewed focus on the development of broadly protective T-cell-based vaccines. Here, we apply structure-based network analysis and assessments of HLA class I peptide stability to define mutationally constrained CD8+ T cell epitopes across the SARS-CoV-2 proteome. Highly networked residues are conserved temporally among circulating variants and sarbecoviruses and disproportionately impair spike pseudotyped lentivirus infectivity when mutated. Evaluation of HLA class I stabilizing activity for 18 globally prevalent alleles identifies CD8+ T cell epitopes within highly networked regions with limited mutational frequencies in circulating SARS-CoV-2 variants and deep-sequenced primary isolates. Moreover, these epitopes elicit demonstrable CD8+ T cell reactivity in convalescent individuals but reduced recognition in recipients of mRNA-based vaccines. These data thereby elucidate key mutationally constrained regions and immunogenic epitopes in the SARS-CoV-2 proteome for a global T-cell-based vaccine against emerging variants and SARS-like coronaviruses.

China Insists The US "Pay Its Share" In Afghan Reconstruction While "Reflecting On Failure"

China is pledging support to the Taliban government now in charge of Afghanistan for reconstruction of the country, but is continuing to add insult to injury in the wake of the disastrous US evacuation and pullout from Kabul of the last two weeks.

"China has pledged to help reconstruction efforts in Afghanistan after American troops have completely withdrawn, but demands that Washington also pay its share," The South China Morning Post writes in a Tuesday report. China's foreign ministry went so far as to demand that Washington "reflect on its failure" after closing down its longest running war in history.

Foreign Ministry spokesman Wang Wenbin issued scathing criticism during a press briefing Tuesday, saying the United States' starting the war in Afghanistan in the first place is ultimately "the reason for public livelihood and economic difficulty in the nation."

"The US has to take up responsibility and cannot just leave the chaos behind," Wang emphasized. "The US has to work with the international community to provide economic and humanitarian assistance to Afghanistan, maintain the normal operations of the government, maintain social stability, stop the currency depreciation and inflation, and let Afghanistan go on the path of peace," he said.

Alternately he touted China's reconstruction efforts as part of a new 'peaceful start' for Afghanistan: "China will support the peaceful reconstruction of Afghanistan on the basis of respecting the wishes and demands of Afghanistan."

The stinging rebuke was laced with repeat comments on Washington's need to 'learn its lesson' in the failure of the Afghan war:

Taliban Move To Ban Opium Production, But Could It Majorly Backfire?

The Taliban have vowed to reduce Afghanistan’s opium trade, according to a report by the WSJ.

This is incredibly suspect, as the movement’s primary bankrolling comes from poppy growing and opium production.

The Islamic group’s spokesman Zabihullah Mujahid vowed to crack down on the production of narcotics, saying “nobody can be involved” in the heroin trade.

Taliban leaders have been telling farmers in the southern province of Kandahar to stop cultivating opium poppies, according to the WSJ report.

Farmers are unhappy but have no choice but to comply should the Taliban begin to enforce the ban, the outlet cites a Kandahar grower as saying.

“We can’t oppose the Taliban’s decision. They are the government,” said the farmer.

He added that the Taliban has assured people that they would have an “alternative crop,” such as saffron, to grow.

Saffron, however, is not nearly as lucrative as producing as producing narcotics.

The ban on a crop that has traditionally been a crucial part of the local economy has resulted in prices of raw opium skyrocketing across the country.

Local farmers in poppy-growing regions like Kandahar, Uruzgan, and Helman provinces said raw opium prices have tripled, from about $70 to about $200 per kilogram. In the northern city of Mazar-e-Sharif, the price of opium has doubled, according to locals.

The poppy-planting season is due start in about a month.

“If the Taliban prohibit the cultivation of poppy, people will die from starvation, especially when international aid stops. We still hope they will let us grow poppies. Nothing can compensate for the income we get from growing poppies,” a poppy farmer in the Chora district of Uruzgan was quoted as saying.

Afghanistan is the world’s leading producer of opium, with its share in the global market standing at over 80%.

Poppy cultivation offers the rural population in the war-torn country a much-needed lifeline. In 2017, annual opium production was valued at $1.4 billion, or 7.4 percent of Afghanistan’s gross domestic product, according to the UN.

The Taliban have used taxes on the drug business to bankroll they endeavors for a while. After seizing the country’s capital Kabul, the issue of the narcotics trade surfaced amid the Taliban’s new plans for governance.

“We are assuring our countrymen and women and the international community, we will not have any narcotics produced. From now on, nobody’s going to get involved (in the heroin trade), nobody can be involved in drug smuggling,” Taliban Spokesman Zabihullah Mujahid told reporters in Kabul at an August 18th press conference.

Before 2001, when the US invaded, Taliban had banned opium production. Production was down by 90%.

The Taliban eventually ceased to mete out punishment for cultivating drugs, cracking down only on use of drugs.

After 2001, during the two decades of deployment of Western forces in the country, the US spent some $9 billion in a crackdown on the drug trade. US efforts included paying farmers to destroy their poppies, funding Afghan eradication teams, and urging people to grow saffron, pistachios, or pomegranates instead.

This, however, pushed many of the local population to join the Taliban’s ranks.

The ban on the opium growing is a risky move, as the country is in an economic crisis, and a precarious one at that.

The US froze Afghanistan’s central bank assets and foreign aid, as well as the local currency – the afghani is reeling, on the brink of collapse.

New Chinese Law Requires Foreign Vessels to Report to Maritime Authorities When Entering Waters

China has long struggled to exert control over the large parts of the South China Sea it claims, including by erecting floating radar stations and dredging the seafloor to expand the waterway’s tiny islands. However, so have several other nations, many of which claim the same islands and waters.

Foreign vessels entering Chinese territorial waters will be required to report their ship and cargo information to the country’s maritime authorities if they’re carrying hazardous cargo or pose a threat to maritime traffic safety beginning on September 1.

The new regulation is the product of revisions to the Maritime Traffic Safety Law made in April by the Standing Committee of the National People’s Congress, a legislative body that remains in session while the larger congress is not.

According to the South China Morning Post, which viewed the agency’s recent notice on the changes, affected vessels include ships carrying radioactive material, bulk oil, chemical, liquefied natural gas and other toxic and harmful substances, as well as to all submersibles, nuclear-powered vessels, or vessels “considered a threat to the country’s maritime traffic safety.”

The information they will have to supply include the vessel’s name, call sign, position, and the dangerous goods on board. Voice of America reported in April that the fine for violation is up to $47,000.

A separate provision that also enters force on Wednesday requires captains to implement “emergency response measures” if a crew member or passenger on their vessel is found or suspected of having an infectious disease that could pose a serious threat to the health of others. The law requires them to be put under quarantine and for the situation to be reported to maritime authorities, according to Xinhua News Agency.

As the new regulation would apply to China’s claimed islands in the South China Sea, many have suggested it is aimed at further solidifying their control over the islands and the waters around them. While the islands themselves are often little more than specks of land in the waterway, a nation’s territorial waters extend 12 nautical miles from shore.

Kang Lin, a deputy director of China’s National Institute for South China Sea Studies, told the SCMP the intention was to “close loopholes” that allowed civilian ships being used for military purposes to evade regulation while in Chinese waters.

“We used to put a lot of effort on the management of warships, while ignoring the supervision and management of civilian commercial ships which are under the cover of civilian commercial use, but in fact engaging in militarised intelligence collection,” Kang said.

In April, Chinese fishermen found a small solar-powered drone vessel packed with antennae and surveillance equipment, which China Central Television described as “a reconnaissance device secretly placed by a foreign country in China’s waters.” The three-meter-long device was not publicly identified, but SCMP noted at the time that many on social media believed it looked very similar to a device built by Liquid Robotics, a subsidiary of American defense contracting giant Boeing.

The law would also affect the Taiwan Strait, which China claims as internal waters because it also claims Taiwan itself as part of China, regarding the government in Taipei as a rebel movement. Taipei Times claimed on Tuesday the regulation is a “ticking time bomb” for conflict in the South China Sea, but said it was likely to have little effect on the strait, as the United Nations Convention on the Law of the Sea regards it as an international strait through which “continuous and expeditious” passage is guaranteed.

The Australian government has been moving towards a surveillance state for some years already. Now they are putting the nail in the coffin with an unprecedented surveillance bill that allows the police to hack your device, collect or delete your data, and take over your social media accounts; without sufficient safeguards to prevent abuse of these new powers.

This month the Australian government has passed a sweeping surveillance bill, worse than any similar legislation in any other five eye country.

The Surveillance Legislation Amendment (Identify and Disrupt) Bill 2020 gives the Australian Federal Police (AFP) and the Australian Criminal Intelligence Commission (ACIC) three new powers for dealing with online crime:

Data disruption warrant: gives the police the ability to "disrupt data" by modifying, copying, adding, or deleting it.

Network activity warrant: allows the police to collect intelligence from devices or networks that are used, or likely to be used, by those subject to the warrant

Account takeover warrant: allows the police to take control of an online account (e.g. social media) for the purposes of gathering information for an investigation.

The two Australian law enforcement bodies AFP and ACIC will soon have the power to modify, add, copy, or delete your data should you become a suspect in the investigation of a serious crime.

What makes this legislation even worse is that there is no judicial oversight. A data disruption or network activity warrant could be issued by a member of the Administrative Appeals Tribunal, a judge's warrant is not needed.

Australian companies obliged to comply

When presented with such warrant from the Administrative Appeals Tribunal, Australian companies, system administrators etc. must comply, and actively help the police to modify, add, copy, or delete the data of a person under investigation. Refusing to comply could have one end up in jail for up to ten years, according to the new bill.

Required hacking activities could include: altering, copying and deleting data; intercepting and modifying communications; surveilling networks; and changing account credentials.

Justification of the bill

Politicians justify the need for the bill by stating that it is intended to fight child exploitation (CSAM) and terrorism. However, the bill itself enables law enforcement to investigate any "serious Commonwealth offence" or "serious State offence that has a federal aspect".

In fact, this wording enables the police to investigate any offence which is punishable by imprisonment of at least three years, including terrorism, sharing child abuse material, violence, acts of piracy, bankruptcy and company violations, and tax evasion.

For lawyer Angus Murray, Chair of Electronic Frontiers Australia’s Policy Team, the hacking powers pose a serious risk to our civil liberties.

"This is now a regime in Australia where we have conferred power on law enforcement agencies to hack Australians’, and potentially overseas persons’, computers and to take over accounts and modify and delete data on those accounts," he told Information Age.

"Australia doesn’t have constitutionally enshrined rights to political speech and other human rights, but if we’re going to give law enforcement these powers, that should be checked and balanced against a human rights instrument at Federal level."

Murray warns that there could come a point where this power is used against society. In theory, at least, the police could put something like child exploitation images onto your computer. While something like this is not the intention of the bill, there are also no significant safeguards against it.
