"The Pfizer RNA based COVID-19 vaccine was approved by the US FDA under an emergency use authorization without long term safety data. Because of concerns about the safety of this vaccine a study was performed to determine if the vaccine could potentially induce prion based disease."

ABSTRACT

Development of new vaccine technology has been plagued with problems in the past. The current RNA based SARS-CoV-2 vaccines were approved in the US using an emergency order without extensive long term safety testing. In this paper the Pfizer COVID-19 vaccine was evaluated for the potential to induce prion-based disease in vaccine recipients. The RNA sequence of the vaccine as well as the spike protein target interaction were analyzed for the potential to convert intracellular RNA binding proteins TAR DNA binding protein (TDP-43) and Fused in Sarcoma (FUS) into their pathologic prion conformations. The results indicate that the vaccine RNA has specific sequences that may induce TDP-43 and FUS to fold into their pathologic prion confirmations. In the current analysis a total of sixteen UG tandem repeats (ΨGΨG) were identified and additional UG (ΨG) rich sequences were identified. Two GGΨA sequences were found. Potential G Quadruplex sequences are possibly present but a more sophisticated computer program is needed to verify these. Furthermore, the spike protein, created by the translation of the vaccine RNA, binds angiotensin converting enzyme 2 (ACE2), a zinc containing enzyme. This interaction has the potential to increase intracellular zinc. Zinc ions have been shown to cause the transformation of TDP-43 to its pathologic prion configuration. The folding of TDP-43 and FUS into their pathologic prion confirmations is known to cause ALS, front temporal lobar degeneration, Alzheimer’s disease and other neurological degenerative diseases. The enclosed finding as well as additional potential risks leads the author to believe that regulatory approval of the RNA based vaccines for SARS-CoV-2 was premature and that the vaccine may cause much more harm than benefit.

NYT: Swiss Billionaire Behind ‘Hub Project’ to Influence U.S. Media, Politics; Bids for Tribune Co.

Swiss billionaire Hansjörg Wyss, who emerged Tuesday as a top bidder for the Tribune Media Company, has also created a group called the Hub Project to influence U.S. elections, primarily through favorable media coverage for Democrats, the New York Times reports.

Wyss, 85, “also sits on the board of directors of the left-wing Center For American Progress, founded by longtime Clinton confidant and 2016 presidential campaign chairman John Podesta,” Fox News reported Tuesday. He also donated millions of dollars to the Clinton Foundation.

The Times reports that Wyss (pronounced Veese) is behind the Hub Project, which was first exposed by Wikileaks in 2016:

The organization, called The Hub Project, was started in 2015 by one of Mr. Wyss’s charitable organizations, the Wyss Foundation, partly to shape media coverage to help Democratic causes. It now has 60 employees, according to its website, including political organizers, researchers and communications specialists. Mr. Wyss and his charitable foundation are not mentioned on The Hub Project’s website, and his role in its creation has not been previously reported.

New coronavirus vaccines defined

Pfizer and BioNTech

Pfizer and its German-based partner BioNTech use a new approach to making vaccines that uses messenger RNA or mRNA.

This design was chosen for a pandemic vaccine years ago because it's one that lends itself to quick turnaround. All that is needed is the genetic sequence of the virus causing the pandemic. Vaccine makers don't even need the virus itself -- just the sequence.

In this case, BioNTech researchers used a little piece of genetic material coding for a piece of the spike protein -- the structure that adorns the surface of the coronavirus, giving it that studded appearance.

Messenger RNA is a single strand of the genetic code that cells can "read" and use to make a protein. In the case of this vaccine, the mRNA instructs cells in the body to make the particular piece of the virus's spike protein. Then the immune system sees it, recognizes it as foreign and is prepared to attack when actual infection occurs.

MRNA is very fragile so it's encased in lipid nanoparticles -- a coating of a buttery substance that can melt at room temperature. That's why Pfizer's vaccine must be kept at ultracold temperatures of about minus 100 degrees F (minus 75 degrees C). That means special equipment is needed to transport and store this vaccine.

Moderna

Moderna's vaccine is also based on mRNA. "mRNA is like software for the cell" Moderna said on its website.

And like the Pfizer/BioNTech vaccine, it codes for cells to make a piece of the spike protein. That was a careful choice -- scientists had to pick a piece of the virus they thought would not mutate, or change much, as time passed. The virus uses the spike protein to grapple the cells it attacks and the structure does appear to stay stable through generation after generation of viral replication.

Moderna has come up with a different formulation for the lipid nanoparticles to protect the mRNA in its vaccine. These formulations are corporate secrets, but Moderna thinks its approach is better and said its vaccine can be shipped at minus 20 degrees C (minus 4 degrees F) and can be kept stable for 30 days at 2 degrees to 8 degrees C (36 to 46F), the temperature of a standard home refrigerator.

AstraZeneca

AstraZeneca's vaccine, made with a team at Britain's Oxford University, is called a vector vaccine. It uses a common cold virus called an adenovirus to carry the spike protein from the coronavirus into cells.

It also aims to make people's bodies in essence produce their own vaccines by churning out little copies of spike protein, but the delivery method is different. This adenovirus infects chimpanzees but doesn't make people sick. It was modified so that it doesn't replicate itself -- then genetically engineered to inject cells with the DNA encoding for the full coronavirus spike protein.

It's a cheaper way to make vaccines -- but slower than using RNA. The company has pledged to make its vaccine available inexpensively to countries around the world. The vaccine can be kept stable for six months at standard refrigerator temperatures, the company said.

Johnson & Johnson's vaccine arm Janssen Pharmaceuticals

Janssen's coronavirus vaccine is a recombinant vector vaccine. Like AstraZeneca's, it uses an adenovirus but this one does infect humans. It is a genetically engineered version of adenovirus 26, which can cause the common cold but the gene tinkering has disabled it. It also delivers the genetic instructions to make spike protein.

This is one vaccine that has been tested on the market before. The adenovirus 26 vector was used to make the company's Ebola vaccine, which won marketing authorization from the European Commission in July.

It's a one-shot vaccine but earlier this month Johnson & Johnson started a two-dose, Phase 3 trial in Britain, because there's some evidence that two doses provide better protection. Volunteers will get two shots, 57 days apart, or placebos.

Novavax

Maryland-based biotechnology company Novavax specializes in "protein subunit" vaccines. They use virus-like nanoparticles as a base and cover them with genetically engineered pieces of the coronavirus spike protein.

This is also a tried and true vaccine approach. A hepatitis B vaccine given to newborns is a protein subunit vaccine, as is the human papillomavirus or HPV vaccine and FluBlok, Sanofi's influenza vaccine.

Novavax uses an insect virus called a baculovirus to get the coronavirus spike protein into moth cells, which then produce the protein. This is harvested and mixed with an adjuvant an immune booster based on saponin, found in soap bark trees.

Sanofi and GlaxoSmithKline

This is also a protein subunit vaccine, using Sanofi's FluBlok technology with a GlaxoSmithKline adjuvant. It also uses a baculovirus to grow little bits of spike protein.

Sinovac and Sinopharm

Chinese company Sinovac's CoronaVac uses an inactivated virus -- one of the oldest methods for vaccinating people. Whole batches of coronavirus are grown, "killed" and then made into vaccine. Likewise, Sinopharm's vaccine in an inactivated virus.

Sputnik V

Russia's Sputnik V coronavirus vaccine is an adenoviral vector vaccine. It uses two common cold viruses called adenovirus 5 and adenovirus 26 to carry the genetic material for the spike protein into the body.

A district court upheld an Ohio law banning abortions of babies diagnosed with Down syndrome Tuesday, marking a victory for the pro-life movement at the state level.

The court found the law did not create an "undue burden" on the right to an abortion because it limited doctors from performing abortions only when a mother discloses she's requesting the procedure because her child has been diagnosed with Down syndrome.

"[The law] furthers three valid and legitimate interests by protecting: (1) the Down syndrome community from the stigma associated with the practice of Down-syndrome-selective abortions, (2) pregnant women and their families from coercion by doctors who advocate abortion of Down-syndrome-afflicted fetuses, and (3) the integrity and ethics of the medical profession by preventing doctors from becoming witting participants in Down-syndrome-selective abortions," the court wrote in its opinion. "[The law] allows doctors to perform such abortions when they do not know that Down syndrome is the reason, without undermining [the law's] specific purposes or objectives."

The court's decision comes in the middle of a number of state laws seeking to restrict access to abortion. Arkansas's attorney general asked the Supreme Court to intervene to reverse the striking down of a similar Arkansas law banning Down-syndrome-selective abortions. Meanwhile, blue state attorneys general are taking action to prevent laws restricting access to abortion from going into effect. New York attorney general Letitia James (D.) led a group of 20 state attorneys general in challenging a Tennessee law requiring women to get two in-person appointments before obtaining an abortion.

Chuck Donovan, president of the pro-life Charlotte Lozier Institute, said state laws restricting abortions for reasons of Down-syndrome diagnoses are pushing the conservative-leaning Supreme Court toward revisiting Roe v. Wade. He said the Court has signaled even if Roe is not overturned, justices are open to chipping away at current law.

"In certain countries, children with Down syndrome are being devastatingly eliminated from the population," Donovan said. "Justice Clarence Thomas has confirmed this is an issue of first impression and it remains an ‘open question' whether states may implement protections such as these. We are encouraged by this ruling and hope the Supreme Court will now weigh in on this important human rights issue."

The Sixth Circuit's decision marks a reversal from the Seventh Circuit appeals court's decision in 2018 to overturn an Indiana law that prohibited abortions based on the gender, race, or disability of a fetus. The overturning of the three judge panel's ruling also makes further review of Ohio's law more likely.

As the Biden administration expands federal support for access to abortion, Republican-controlled states are appealing to the judiciary. The likelihood of further review of the Ohio law, combined with Arkansas's direct appeal to the Supreme Court and blue state attorneys general challenging laws restricting abortion, forecasts a judicial showdown over abortion could be imminent.

Biden DOJ Pick Struggles to Defend Record on Black Panther Prosecutions

During confirmation hearing, Republicans pushed Kristen Clarke on her history of opposing civil rights cases against black defendants

Justice Department nominee Kristen Clarke struggled to explain her criticisms of a civil rights case lodged against members of the New Black Panther Party during her confirmation hearing Wednesday.

Clarke tried to duck questions about her opposition to prosecuting two Panthers for civil rights violations in the 2008 election while testifying before the Senate Judiciary Committee. Republican lawmakers say the Black Panther case is a troubling indicator of her approach to enforcing civil rights laws.

Clarke's history of opposing civil rights prosecutions of black defendants has made her nomination contentious. If confirmed, she will lead the Justice Department's Civil Rights Division and serve as top prosecutor for a range of nondiscrimination statutes.

Sen. Mike Lee (R., Utah) asked Clarke directly if the Justice Department was wrong to pursue a case against the Panthers. Clarke dodged, saying the department has the prerogative to bring any cases it deems appropriate. Lee doubled down, asking if she wanted prosecutors to abandon the case.

"I may have espoused that opinion publicly, but so did many people," Clarke replied. "I think there were many people who deemed that a weak case."

Florida House passes contentious transgender sports ban

TALLAHASSEE — The GOP-controlled Florida House on Wednesday passed controversial legislation banning transgender athletes from playing girls’ sports, shifting attention to the state Senate where final approval is needed to send the bill to Gov. Ron DeSantis.

The 77-40 mostly party-line vote, with all but one Democrat opposing, came one day after House Democrats held up a floor session for hours attempting to scale back the measure. GOP lawmakers say the bill is needed to protect the sanctity of women’s sports, but opponents contend it would only “legalize bullying” for transgender students.

“HB 1475 is purely political, and it plays on the fears and the ignorance about the transgender community in order to score partisan points,” state Rep. Carlos Guillermo Smith (D-Orlando), an openly gay lawmaker, told reporters ahead of Wednesday’s vote.

House and Senate Republicans in Florida have seized on women’s sports as a priority in 2021, following the path of more than 20 other GOP-leaning states that are using the issue to limit transgender rights. LGBTQ advocacy groups like Equality Florida are aligned with the majority of Democrats in opposing the legislation, arguing it would lead to increased stigma and misinformation surrounding transgender students.

The idea issue was magnified on Monday when the NCAA put states like Florida on notice, warning that locations that don’t treat all student athletes with “dignity and respect” could be ineligible to host future championship games.

West Virginia Gov. Jim Justice reiterated Wednesday that he doesn’t plan to veto a bill banning transgender middle school, high school and college girls and women from playing on women’s sports teams, even though he conceded it could lead the NCAA to “penalize us.”

The NCAA said in a news release Monday that it might no longer hold college championships in states with such laws. The legislation (House Bill 3293) would implement the ban only at public schools, colleges and universities.

“Even at our college level, I support the bill there as well,” Justice said. “The only problem is just one thing, and that is this: The NCAA, you know, has moved to a more political, politically correct or liberal body. And they could very well penalize us.”

Second UK inquiry into ex-PM David Cameron lobbying scandal announced by select committee of MPs

The UK's Treasury Committee has announced it will hold an inquiry into the government lobbying scandal surrounding former Prime Minister David Cameron, which has now engulfed several ministers and is already facing a review.

The cross-party committee of MPs, which examines Treasury expenditure, administration and policy, said in a statement on Wednesday it has agreed “in principle” to an inquiry, which will be launched next week.

The group said their investigation will be into the “lessons” from the scandal, which involves collapsed financial lender Greensill Capital as well as former Tory PM Cameron.

Prime Minister Boris Johnson on Monday announced an independent review by legal expert Nigel Boardman into Greensill and the use of supply chain finance in government, although he did not explicitly mention lobbying.

Earlier on Wednesday, Johnson's Conservative government voted down a plan by the opposition Labour party to hold a wider parliamentary inquiry into the lobbying.

Labour leader Sir Keir Starmer said he wanted a “full” investigation into the scandal, which he said had exposed the “sleaze and cronyism” at the “heart” of the Conservative Party.

Cameron, who became an adviser for Greensill after he left Downing Street, allegedly lobbied ministers on its behalf in a bid to win government contracts for the firm.

His actions included sending text messages to Chancellor Rishi Sunak and taking Lex Greensill, the company's founder, for a private drink with Health Secretary Matt Hancock.
