As far as specific suggestions, if you don't know what layers, layer masks, cloning, and selections can do, they are your core, but know what your chosen editor is capable of and be creative. Above all else, practice. I didn't learn anything overnight. Also be persistent. Harder edits will frustrate the hell out of you but trying hard enough usually will usually let you come up with something workable.

First, I remember having an image with most of the fonts used previously (I think I got it from the bunker), so there should be no problem in that part.
Second, how faithful should I be with the font used in titles? Is pic related okay?

Looking at previous chapters, there seems to be a number of different fonts used to render the titles in English. Might be worth trying to check what those are, as well as if the Japanese titles had similar variety of font or not (and thus if there's any pattern to what the previous English titles have used). I guess the real concern is that it fits with what's established.
As to the example though: I kind of think a less "bubbly" font for the "Despair" line might work better as the Japanese script isn't especially thick, but definitely keep a handwritten look to it. I'd also say that you ought to try it as white with no stroke if you're going to be using shadowing around it to pop it from the background (that effectively gives it a light stroke on its own). At the same time, the shadowing feels a bit too thick in both cases: Notice how easily quickly it dissipates on the Japanese. Just my thoughts though.

Well, it's less thick, but I'm still not sure it's creating a smoothly fading gradation.
Try this, since you're using photoshop: Set the foreground color to black, and then after selecting the text layer, open the option to add an outer glow. In the gradient option, select Foreground-to-Transparent from the options, adjust the size so it feels at how with how much the effect extends on the Japanese render, and diffuse the effect via the noise slider. Might take some playing around with Pic related as a quick sample (ignore the font).

Oh, and one more thing: First thing you should do in the outer glow options is set the blend mode to darken, or something of that nature. Program default is to use the effect to lighten (it is called a glow, after all), which obviously won't be useful when the point is to pop white text against a white background. Otherwise you wouldn't be able to see it.

Again, I'd say to play with the formatting of some of the bubbles throughout the chapter, but especially the thoughts in panel 2 there. It feels really cramped to me. I don't know that the exact font or size you're using there is, and only have a basic estimate of the room to work with, so pic related might not be the best example, but some better vertical distribution might help with dealing with the horizontal space. Also, I think it needs some fixes with commas and spelling, though that could well be on the transcript.
You won't always be able to fit the framing of the bubbles as best you'd like, but do it where you can, or at least make it feel well balanced (and if extending past the bubble, stroke the text).
