The Importance of microRNAs in RAS Oncogenic Activation in Human Cancer

Introduction
microRNAs (miRNAs) are small (19–24 nucleotides) non-coding RNAs discovered in 1993 in studies related to embryonic development of C. elegans (1, 2). Their importance significantly increased following the discovery of their existence in all eukaryotic organisms (3). Currently, 2,654 mature miRNAs, originating from 1917 precursors, are described in humans (http://www.mirbase.org/) (4, 5). Their main function is to negatively regulate gene expression at the post-transcriptional level through the interaction of their “seed” portion by sequence homology typically with the 3′ non-coding regions of messenger RNAs (mRNAs). Through this interaction, miRNAs limit translation, or promote degradation of target mRNAs (6, 7).
The modulation of target mRNAs by miRNAs is complex, considering that each mRNA is generally targeted by multiple miRNAs, and the strength of this interaction is variable (8). Classically, it has been thought that each miRNA can interact with hundreds of target mRNAs. However, recent reports have highlighted RNA transcripts inducing degradation of respective interacting miRNAs through a mechanism known as “target-directed miRNA degradation” (TDMD) (9, 10). Added to the complexity of these direct interactions is the fact that some long non-coding RNA (lncRNA) could function as “sponges,” that act as a buffer and prevent the action of miRNAs on target protein-coding mRNAs (11, 12). Lastly, it is also important to consider that cell co-localization of each miRNA with the target mRNAs is necessary and depends on the eventual tissue-specific expression of each of the interacting RNAs.
Thus, miRNAs, taken together, represent an essential phase in the regulation of gene expression by modulating the translation of the entire transcriptome (13, 14). Given their biological importance, their deregulation plays a significant role in pathogenic mechanisms, including the neoplastic transformation (15, 16). The first evidence associating miRNAs with human malignant diseases was the discovery of miR-15 and miR-16 in the minimal region of deletion at chromosome 13q14 in chronic lymphatic leukemia (17). Since this seminal study, a myriad of other studies has confirmed the role of miRNAs in tumorigenesis and other human diseases as well.
Conclusions
Taken together, published data provides a strong indication that altered miRNA expression represents an important mechanism for RAS activation, with various implications. First, it represents a mechanism of pathogenic relevance, responsible for the promotion of several tumor traits, irrespective of RAS oncogenic mutations. Second, considering that the activation of RAS represents a frequent mechanism of resistance for drugs directed against RTKs, it is possible that miRNA dysregulation represents a relevant aspect to consider when assessing the proper management of patients on target therapies. Third, miRNAs may represent potentially useful molecules for the control of RAS oncogenic activation, aimed at overcoming the lack of drugs targeting RAS and possibly improving the efficacy of target therapies.

The world’s number one ranked tennis player Novak Djokovic was deported from Australia on Saturday and banned from entry for the next three years.

The UK government admits that vaccines have damaged the natural immune system of those who have been double-vaccinated.
The UK government has admitted that once you have been double-vaccinated, you will never again be able to acquire full natural immunity to Covid variants – or possibly any other virus.
In its Week 42 “COVID-19 Vaccine Surveillance Report”, the UK Department of Health admits on page 23 that “N antibody levels appear to be lower in people who become infected after two doses of vaccination”.
It goes on to say that this drop in antibodies is essentially permanent.
vaccinated people do not appear to form antibodies against the nucleocapsid protein, the envelope of the virus, which is a crucial part of the response in unvaccinated people.
In the long term, the vaccinated are far more susceptible to any mutations in the spike protein, even if they have already been infected and cured once or more.
The unvaccinated, on the other hand, will gain lasting, if not permanent, immunity to all strains of the alleged virus after being naturally infected with it even once.

Republican senators find NIAID (National Institute of Allergy and Infectious Diseases) Director, Dr. Anthony Fauci has questionable investments in Chinese-tied companies.
On Saturday, Sen. Roger Marshall (R-Kan.) released Fauci’s financial disclosures showing his investments in the Matthews Pacific Tiger Fund among other things. That fund has nearly 43 percent of its portfolio invested in Hong Kong and China, including a Chinese pharmaceutical company.
Overall, Fauci has almost $10.5 million invested in various companies and critics are sounding the alarm over his potential conflicts of interests. Judicial Watch President, Tom Fitton specially commented on the matter.

Clinton appeared as a passenger on Epstein's flight logs at least 26 times in 2002 and 2003. Epstein's plane was known then as the "Lolita Express."
Jeffrey Epstein's attorney contacted the couple who managed Jeffrey Epstein's private island in 2016 to request that they sign an affidavit saying that former-President Bill Clinton did not visit Little St. James, Epstein's private island.
According to the Daily Mail, lawyer Darren Indyke contacted the couple, Miles and Caty Alexander, asking them to state that Clinton had not been on seen on the island, and that Clinton had not been on the island while the couple was absent from the island.
“It was during the 2016 presidential campaign which was won by Trump. There was a worry that the link between Epstein and Clinton and in particular the island could be used against Hillary," a source told the Mail.
Official flight logs from the White House during the former president's first term show that Clinton visited the island at least 17 times from 1993 and 1995.
Additionally, Clinton appeared as a passenger on Epstein's flight logs at least 26 times in 2002 and 2003. Epstein's plane was known then as the "Lolita Express."
While Clinton denies ever being on the island, alleged Epstein victim Virginia Giuffre claims that she saw Clinton there.
The two appear to have been photographed together, with one picture showing Giuffre giving the president a shoulder rub.
Giuffre has also said that was Clinton on the island in the company of two "young girls."

Maxwell’s legal team revealed she will no longer protect the names of “eight John Does” who raped young children given to them by Jeffrey Epstein.
One of Maxwell’s attorneys stated in a letter to federal Judge Loretta Preska this week that she would no longer protect the identities of the high-profile “John Does.”
The names were sealed in a 2015 civil lawsuit brought against Maxwell by Prince Andrew’s child rape victim Virginia Roberts Giuffre.
Maxwell had previously refused to identify the VIP pedophiles.

Just want to say thank you for all the kind posts last bread, ate some spicy raman a little endorphins. I just wish i could do more physical things like one anon suggest running but i lost a bunch of my mobility a few years ago and last year i could not ride my scooter any more due to my mobility got even worse. Trying to lift some weights and i am happiest drawing so i will just drown myself in that and fight the culture the best way i can with my art and to make comics and stories good again. The US military is poisoning the water in O’ahu
Native Hawaiians have been leading a drawn-out fight against the US military’s poisoning of the local water supply from leaking petroleum at the Red Hill Fuel Storage Facility in O’ahu.
As Director for the Sierra Club of Hawai’i Wayne Tanaka recently wrote in The Guardian, the US Navy’s Red Hill Fuel Storage Facility is “a massive underground ‘farm’ of 18 million-liter fuel tanks and pipes just 100 feet above metropolitan O’ahu. Its construction began before the Japanese attacked Pearl Harbor. Since then, it has leaked over 180,000 gallons of petroleum into the groundwater aquifer that provides drinking water for over 400,000 residents and visitors from Hālawa to Hawaiʻi Kai.”
Regardless of the major threat the facility poses to the local water system and demands from Native Hawaiians and supporters to address the crisis and hold the US military accountable, it wasn’t until hundreds of military families living near Pearl Harbor reported symptoms of petroleum poisoning that Red Hill’s operations were paused in late November. But the root causes of the environmental and public health crisis remain untouched, and the fight to shut down Red Hill is still very much ongoing.
In this segment of The Marc Steiner Show, Marc speaks about that fight with Mikey Inouye, an independent filmmaker born and raised in Hawai‘i, community organizer, and member of O‘ahu Water Protectors. The O‘ahu Water Protectors is an organization that formed out of a coalition of Kānaka Maoli organizers, Sierra Club members and supporters, Hawai‘i Peace and Justice, and other groups working toward sovereignty, decolonization, and demilitarization.
