Should there be more series with bio-organic mechas? Should there even be a series with bio-organic mecha that is equal parts horror and action? Should we even have a series that treats these kinds of bio-machines with the Lovecraftian mysticism the Demon of Steel OVA treats them?

And many more questions. So far, the only things that fit close the bill, from memory, are of course the Einst from Banpresto and the Bird Human from Macross…can't remember if the Gilgamesh units from Macross II fit that bill. Surprisingly, the red Gilgamesh unit from Macross II bears a very eery resemblance to the red mecha from the HnO movie, the titular Hagane. Right down to the two thumb hands.

The weirdest thing of all is that I haven't found games with them as the main mechas, just as enemies only. Could they work in a Zone of the Enders type of game or some other type?

Evangelions are organic/cybernetic under their armor plating. But that's all I can really think of for organic mecha games.

For the West, I think Warhammer the Orks use huge kaiju animals for their Baneblade equivalents, but I forget the name.

There aren't many media that focuses on organic mecha because when most people think of giant robots they think machines. Even if humans are basically organic mecha anyways.
