Researchers aim to develop edible plant-based mRNA vaccines
The future of vaccines may look more like eating a salad than getting a shot in the arm. UC Riverside scientists are studying whether they can turn edible plants like lettuce into mRNA vaccine factories.
Messenger RNA or mRNA technology, used in COVID-19 vaccines, works by teaching our cells to recognize and protect us against infectious diseases.
One of the challenges with this new technology is that it must be kept cold to maintain stability during transport and storage. If this new project is successful, plant-based mRNA vaccines -; which can be eaten -; could overcome this challenge with the ability to be stored at room temperature.
The project's goals, made possible by a $500,000 grant from the National Science Foundation, are threefold: showing that DNA containing the mRNA vaccines can be successfully delivered into the part of plant cells where it will replicate, demonstrating the plants can produce enough mRNA to rival a traditional shot, and finally, determining the right dosage.
"Ideally, a single plant would produce enough mRNA to vaccinate a single person," said Juan Pablo Giraldo, an associate professor in UCR's Department of Botany and Plant Sciences who is leading the research, done in collaboration with scientists from UC San Diego and Carnegie Mellon University.
We are testing this approach with spinach and lettuce and have long-term goals of people growing it in their own gardens. Farmers could also eventually grow entire fields of it."
Juan Pablo Giraldo, Associate Professor, Department of Botany and Plant Sciences, University of California - Riverside
Key to making this work are chloroplasts -; small organs in plant cells that convert sunlight into energy the plant can use. "They're tiny, solar-powered factories that produce sugar and other molecules which allow the plant to grow," Giraldo said. "They're also an untapped source for making desirable molecules."
In the past, Giraldo has shown that it is possible for chloroplasts to express genes that aren't naturally part of the plant. He and his colleagues did this by sending foreign genetic material into plant cells inside a protective casing. Determining the optimal properties of these casings for delivery into plant cells is a specialty of Giraldo's laboratory.
For this project Giraldo teamed up with Nicole Steinmetz, a UC San Diego professor of nanoengineering, to utilize nanotechnologies engineered by her team that will deliver genetic material to the chloroplasts.
"Our idea is to repurpose naturally occurring nanoparticles, namely plant viruses, for gene delivery to the plants," Steinmetz said. "Some engineering goes into this to make the nanoparticles go to the chloroplasts and also to render them non-infectious toward the plants."
For Giraldo, the chance to develop this idea with mRNA is the culmination of a dream. "One of the reasons I started working in nanotechnology was so I could apply it to plants and create new technology solutions. Not just for food, but for high-value products as well, like pharmaceuticals," Giraldo said.
He is also co-leading a related project using nanomaterials to deliver nitrogen, a fertilizer, directly to chloroplasts, where plants need it most.
Nitrogen is limited in the environment, but plants need it to grow. Most farmers apply nitrogen to the soil. As a result, roughly half of it ends up in groundwater, contaminating waterways, causing algae blooms, and interacting with other organisms. It also produces nitrous oxide, another pollutant.
This alternative approach would get nitrogen into the chloroplasts through the leaves and control its release, a much more efficient mode of application that could help farmers and improve the environment.
The National Science Foundation has granted Giraldo and his colleagues $1.6 million to develop this targeted nitrogen delivery technology.
"I'm very excited about all of this research," Giraldo said. "I think it could have a huge impact on peoples' lives."

The vaccines "are not contaminating our food supply," said Melissa Brown, a Northwestern University immunology professor.
We’ve repeatedly debunked claims that vaccines can be transmitted to people by means other than direct injection.
The headline shared on Facebook accompanied an article that started, "This could be one of the most alarming warnings yet from Dr. Malone. He wrote an essay on how mRNA vaccines are being injected into livestock and companion animals. That means, if you consume the vaccinated animal, the mRNA vaccine enters your body."
The essay is a Jan. 11 blog post by Malone, but it doesn’t mention the food supply.
MRNA vaccines help the body produce a viral protein, which triggers the body to produce antibodies to help protect against infection. That’s slightly different from other vaccines, which contain weakened or dead bacteria or virus to trigger an immune response and help prevent infection.

Republicans have spoken out against Pelosi's phrasing, saying that in America, a person is innocent until proven guilty.
Former President Trump has also criticized the grand jury's decision, calling it "Political Persecution and Election Interference at the highest level in history."
Critics have slammed former Speaker of the House Nancy Pelosi after the Democrat tweeted that former President Donald Trump will have a chance to "prove innocence" in court.
Pelosi was responding to the decision of a Manhattan grand jury to indict the former president over his alleged role in hush money payments to adult film star Stormy Daniels in 2016.
It's not yet clear exactly what charges the former president will be facing, but he has denied any wrongdoing—as well as denying having an affair with Daniels—and repeatedly criticized Manhattan District Attorney Alvin Bragg.

A partner at Cohen Milstein Sellers & Toll, a law firm representing the DNC and suing Trump over January 6 on behalf of Democrats in Congress, was arrested for possessing child pornography.
Michael T. Dolce, a lawyer who specialized in fighting child sex abuse, was arrested by the feds at his West Palm Beach apartment on March 15.
Detectives discovered nearly 2,000 images of child sex abuse on Michael Dolce’s devices.
According to the federal criminal complaint, investigators found sex abuse images with girls as young as 5 years old, Local 10 reported.
Mr. Dolce’s LinkedIn listed him as a partner at Cohen Milstein Sellers & Toll PLLC.
According to reports, Michael Dolce is no longer listed on the Cohen Milstein Sellers & Toll website.

Federal prosecutors accused a 53-year-old South Florida attorney who represents victims of child sexual abuse of possessing child sexual abuse material himself.
According to prosecutors, who announced the arrest Wednesday, Michael T. Dolce is expected to appear in a West Palm Beach federal court Thursday where he is facing multiple child pornography charges.
According to his arrest affidavit, on March 15, FBI agents executed a search warrant at Dolce’s West Palm Beach apartment.
Detectives said they discovered Dolce actively downloading child sexual abuse material using peer-to-peer software.
Investigators said nearly 2,000 images and videos of child pornography were recovered from his devices.
According to a federal criminal complaint, the images and videos depicted girls as young as 5 years old.
Investigators said after confiscating Dolce’s phone, it was found unlocked with a background image featuring a female between the ages of 16 and 25 engaged in sexual intercourse with two men.
A spokesperson for the firm confirmed to Local 10 News that Dolce no longer works there.
“The firm is stunned and saddened by these appalling allegations,” the spokesperson said in a statement Wednesday night. “Michael Dolce was terminated and is no longer affiliated with the firm. We are focused on attending to the needs of our clients and staff, and continuing to cooperate fully with the investigation.”

Investigation Reveals 'Revolving Door' Of DOJ, Big Tech Employees
In the midst of an ongoing lawsuit against the Biden administration alleging collaboration with social media companies to censor Americans, a new report has detailed the extent to which former Justice Department (DOJ) employees are now working at “Big Tech” firms.
The American Accountability Foundation (AAF) investigated the resumes of recent hires and found that more than 360 current employees of Google, Amazon, Microsoft, Apple, and Facebook/Meta are former DOJ employees. Likewise, more than 40 DOJ employees, many of them in senior roles, previously worked at Big Tech firms.
According to the report, since President Joe Biden took office, Google hired 40 former DOJ staffers, Amazon hired 61, Microsoft hired 26, and Meta hired 53.
While staff often move between government and the private sector, “this case is different because unlike government and industry moving expertise back and forth (knowledge about procurement rules for example) what we have seen moving back and forth between DOJ and tech is a shared political agenda, specifically to silence conservative voices,” Yitz Friedman, AAF communications manager, told The Epoch Times.
“Sadly, it is evidence that in the Big Tech community, corporate policy is the government’s policy.”
This concern is heightened by recent evidence of collaboration between the DOJ and Twitter to silence Americans, particularly regarding political speech.
After buying social media platform Twitter, Elon Musk released thousands of internal emails that allegedly showed collusion between Twitter and DOJ officials to censor speech, including banning a report by the New York Post before the 2020 election that reportedly incriminated then-candidate Joe Biden in illicit payments scandals.
‘Campaign of Public Threats’
The release of the “Twitter files” follows a lawsuit by Louisiana Attorney General Jeff Landry and former Missouri Attorney General Eric Schmitt that alleges illegal collusion between the Biden administration and Big Tech companies to suppress free speech.
In a November 2022 interview with The Epoch Times, Landry stated that, because of the First Amendment, “the government doesn’t have the ability to censor speech, especially political speech. And so they can’t go out there and force these companies [to do it].”
Plaintiffs in this case filed a 364-page “finding of fact” document showing a “campaign of public threats against social-media platforms to pressure them to censor more speech on social media.” This document will be discussed in a congressional hearing on March 30 on the “Weaponization of the Federal Government.”
