Pompeo isn’t the first intelligence leader in the Trump administration to accuse Schiff of leaking classified information. In January 2021, former acting Director of National Intelligence Richard Grenell claimed Schiff and his team “regularly” leaked classified information.
Sen. Tom Cotton (R-AR) suggested to conservative commentator Lisa Boothe in April 2021 that Schiff could be a source for dubious reports on Russia offering bounties to Afghan militants to kill U.S. troops. Schiff spokesman Patrick Boland shot back with a brief response to the Washington Examiner at the time, “Yet another false and baseless claim by Tom Cotton — you would think he would know better by now.”

The aliens that started engaging me a few years ago claim to be the zetas of nancy lieder's zetatalk.com. At first they led me to believe they wanted me to continue zetatalk after nancy lieder dies. They proceeded to make it clear that zetatalk was just a psyop. Nancy was an unwitting dupe. She talks to these aliens for real, but she is a narcissist who is in love with the idea of being the chosen one, and they use her.
They tell a story of earth apocalypse by means of a poleshift. The only thing that shifts with any consistency though is the goalpost within this narrative. They routinely berate both nancy and their own apocalypse message.
It's impossible for me to know anything for sure of course. What I can say for certain is these aliens are real. They control our reality both at an individual and mass scale. They use telepathy to communicate and to steer people's thoughts and actions.
Their telepathic communication sounds just like someone is talking in the room with you. Their telepathic suggestion doesn't feel like anything at all. You just have random thoughts and impulses as if they are your own, but they aren't random and they aren't your own.

The Pentagon is planning to boost its production of artillery ammunition by 500% over the next two years as the US is depleting its military stockpiles by sending millions of shells to Ukraine, The New York Times reported Tuesday.
Since Russia launched its invasion of Ukraine, the US has pledged to send Ukraine over one million 155mm artillery shells. Before the US Army began efforts to increase production, it produced 14,400 155mm shells a month, but under the new plans, the number could reach over 90,000 each month.

Abstract
Mimicking natural evolution by DNA shuffling is a commonly used method for the optimization of DNA and protein properties. Here, we present an advancement of this approach whereby a gene library is amplified using a standard polymerase chain reaction (PCR), but incorporates dUTP as a fragmentation-defining exchange nucleotide, together with the four standard dNTPs. Incorporated uracil bases are excised using uracil-DNA-glycosylase, and the DNA backbone subsequently is cleaved with piperidine. This oligonucleotide pool is then reassembled with an internal primer extension procedure using a proofreading polymerase to increase yield, and, finally, is amplified by PCR. Denaturing polyacrylamide urea gels demonstrate this method to produce adjustable fragmentation size ranges dependent on the dUTP:dTTP ratios. Using the model protein, chloramphenicol acetyltransferase I, the sequencing of shuffled gene libraries based on a PCR containing 33% dUTP revealed a low mutation rate, of approx 0.1%, with an average parental fragments size of 86 bases, even without the use of a fragment-size separation. Nucleotide exchange and excision technology (NExT) DNA shuffling is, thus, reproducible and easily executed, making it superior to competing techniques. Additionally, NExT fragmentation outcome can be predicted using the computer software, NExTProg.

The government's campaign to fight "misinformation" has expanded to adapt military-grade artificial intelligence once used to silence the Islamic State (ISIS) to quickly identify and censor American dissent on issues like vaccine safety and election integrity, according to grant documents and cyber experts.
The National Science Foundation (NSF) has awarded several million dollars in grants recently to universities and private firms to develop tools eerily similar to those developed in 2011 by the Defense Advanced Research Projects Agency (DARPA) in its Social Media in Strategic Communication (SMISC) program.
DARPA said those tools were used "to help identify misinformation or deception campaigns and counter them with truthful information," beginning with the Arab Spring uprisings in the the Middle East that spawned ISIS over a decade ago.
The initial idea was to track dissidents who were interested in toppling U.S.-friendly regimes or to follow any potentially radical threats by examining political posts on Big Tech platforms.
DARPA set four specific goals for the program:
"Detect, classify, measure and track the (a) formation, development and spread of ideas and concepts (memes), and (b) purposeful or deceptive messaging and misinformation.
Recognize persuasion campaign structures and influence operations across social media sites and communities.
Identify participants and intent, and measure effects of persuasion campaigns.
Counter messaging of detected adversary influence operations."
Mike Benz, executive director of the Foundation for Freedom Online has compiled a report detailing how this technology is being developed to manipulate the speech of Americans via the National Science Foundation (NSF) and other organizations.
"One of the most disturbing aspects of the Convergence Accelerator Track F domestic censorship projects is how similar they are to military-grade social media network censorship and monitoring tools developed by the Pentagon for the counterinsurgency and counterterrorism contexts abroad," reads the report.
"DARPA's been funding an AI network using the science of social media mapping dating back to at least 2011-2012, during the Arab Spring abroad and during the Occupy Wall Street movement here at home," Benz told Just The News. "They then bolstered it during the time of ISIS to identify homegrown ISIS threats in 2014-2015."
The new version of this technology, he added, is openly targeting two groups: Those wary of potential adverse effects from the COVID-19 vaccine and those skeptical of recent U.S. election results.

A California judge issued a preliminary injunction against a state law that empowers the Medical Board of California to discipline physicians who support opinions about COVID-19 that are not in line with the "consensus," according to reports.
The law, known as Assembly Bill 2098, was set to take effect on Jan. 1, 2023. Under the law, the Medical Board of California and the Osteopathic Medical Board of California could discipline physicians who "disseminate" information about COVID that is not in line with the "contemporary scientific consensus."
But in November, a group of five California physicians filed a lawsuit against California Gov. Gavin Newsom’s administration, saying the law violates their First Amendment rights and constitutional right to due process.
Aaron Kheriaty, MD, is listed as one of the physicians in the lawsuit, and on Wednesday he posted on Twitter that a judge granted their request for a preliminary injunction against AB 2098.
The preliminary injunction temporarily halts the implementation of the law while the case is tried in court.

Drag Time Story Hour for Kids Cancelled by French Mayor after Backlash
A French mayor has cancelled a drag queen storytime event for children after backlash and reorientated the event for adults only, after a petition against the event garnered around 5,000 signatures.
Mayor of Toulouse Jean-Luc Moudenc announced that the planned drag queen event, which was planned for February 18th and originally for children between the ages of three and six, would be reoriented so that only adults would be able to attend after facing a backlash.
The announcement came on Tuesday after a petition organised by the group Furie Française, an activist group made up of some former members of the anti-mass migration youth group Generation Identitaire, along with the pro-traditional family movement Manif Pour Tous, Franceinfo reports.
“This choice of programming – which did not give rise to any visa or endorsement from elected officials – can destabilize part of the public,” Mayor Moudenc said and added that it disapproved strongly of various comments made toward the drag performers on social media networks.
The city stated that it “strongly condemns any form of extremism, threat or discriminatory expression towards Shanna Banana and Brandy Snap, who intervene in the workshop in question,” and added that it called on Interior Minister Gerald Damanin to dissolve the Furie Française group, claiming it to be “the origin of the threats and disturbance of public order.”
Darmanin dissolved the Generation Identitaire group in 2021, arguing that the activists in the group had incited violence and accused them of forming a private “militia” and incited hate speech.
The backlash to the drag queen event in Toulouse is not an isolated case in France in recent months, as another show in Bordeaux saw backlash in December, though the local municipal government supported the event.
Other European countries have also seen similar criticisms of drag queen events for children, such as in Sweden, where members of the populist Sweden Democrats spoke out about the events and attempted to have an event cancelled at a library in Trelleborg last month.
Green Party spokeswoman Märta Stenevi reacted to the attempt by comparing the populists to Russian President Vladimir Putin.

Walker says that Directed Evolution is different than Gain-of-Function, which is defined as “a mutation that confers new or enhanced activity on a protein.” In other words, it means that a virus such as COVID can become more potent depending on the mutation / scientific experiment performed on it.
The Pfizer executive told a Veritas journalist about his company’s plan for COVID vaccines, while acknowledging that people would not like this information if it went public.
“One of the things we [Pfizer] are exploring is like, why don't we just mutate it [COVID] ourselves so we could create preemptively develop new vaccines, right? So, we have to do that. If we're gonna do that though, there's a risk of like, as you could imagine no one wants to be having a pharma company mutating f**king viruses,” Walker said.
“From what I’ve heard is they [Pfizer scientists] are optimizing it [COVID mutation process], but they’re going slow because everyone is very cautious – obviously they don’t want to accelerate it too much. I think they are also just trying to do it as an exploratory thing because you obviously don’t want to advertise that you are figuring out future mutations,” he said.
“Don’t tell anyone. Promise you won’t tell anyone. The way it [the experiment] would work is that we put the virus in monkeys, and we successively cause them to keep infecting each other, and we collect serial samples from them.”
Walker drew parallels between this current Pfizer project and what may have happened at the Wuhan Institute of Virology in China.
“You have to be very controlled to make sure that this virus [COVID] that you mutate doesn’t create something that just goes everywhere. Which, I suspect, is the way that the virus started in Wuhan, to be honest. It makes no sense that this virus popped out of nowhere. It’s bullsh*t,” he said.
“You’re not supposed to do Gain-of-Function research with viruses. Regularly not. We can do these selected structure mutations to make them more potent. There is research ongoing about that. I don’t know how that is going to work. There better not be any more outbreaks because Jesus Christ,” he said.

W.H.O. Calls for ‘Immediate Action’ After 300 Child Cough Syrup Deaths in 2022
“These contaminants are toxic chemicals used as industrial solvents and antifreeze agents that can be fatal even taken in small amounts, and should never be found in medicines,” W.H.O. declared.
The primary contaminants cited by W.H.O. were diethylene glycol (DEG) and ethylene glycol (EG). These substances are apparently added to the cough syrups in question as a sweetener, but as W.H.O. pointed out, they are meant to be ingredients in products like antifreeze and polyester. The human body breaks ethylene glycol down into toxic components that can attack the heart, kidneys, and central nervous system.
W.H.O. said most of the victims of the contaminated syrup were children under the age of five. The largest clusters of victims were reported in Gambia, Indonesia, and Uzbekistan. W.H.O. previously issued medical alerts for each of these countries.
The other four countries where suspect child medicines may have been sold are Cambodia, the Philippines, East Timor, and Senegal.
In December, the Health Ministry of Uzbekistan blamed a particular brand of syrup called Doc-1 Max for killing at least 18 children. The syrup is produced by an Indian company called Marion Biotech, which halted production of the Doc-1 Max line pending an inquiry. Laboratory tests performed in Uzbekistan found potentially harmful amounts of ethylene glycol in the medicine.
In early January, W.H.O. advised consumers worldwide to stop using children’s cough syrup from Marion Biotech and another Indian company, Maiden Pharmaceuticals, whose products were implicated in the deaths of Gambian children.
The Indian government suspended Marion Biotech’s production license around the time this W.H.O. advisory was issued. Maiden’s plant was also shut down, but the company is hoping to restart production after a government inspection.
Another W.H.O. alert warned consumers to stop using cough syrup from four Indonesian companies called PT Yarindo Farmatama, PT Universal Pharmaceutical, PT Konimex, and PT AFI Farma.
W.H.O. is reportedly investigating the possibility that the two Indian firms and four Indonesian firms obtained ingredients for their cough syrups from the same suppliers. PT Universal Pharmaceutical, one of the few companies willing to comment on the allegations, pointed to these suppliers as the likely source of the dangerous ingredients.
“Chase the suppliers, they’re the real criminals. They’re the ones that forge raw ingredients by falsifying raw ingredient documents all the way to pharmaceutical companies,” said PT Universal Pharmaceutical lawyer Hermansyah Hutagalung, without naming the “criminals” in question.
“Since these are not isolated incidents, W.H.O. calls on various key stakeholders engaged in the medical supply chain to take immediate and coordinated action,” the Monday bulletin stated.
W.H.O. Acting Director for Antimicrobial Resistance Hanan Balkhy warned at a news conference on Tuesday that “there might be children out there exposed to these medications that we’re not even aware of.”

The Nov. 1 death of Daniel Best, a pharmaceutical executive from Bay Village who led U.S. Department of Health and Human Services efforts to lower prescription drug prices, has been ruled a suicide, officials in Washington, D.C., said Thursday.
Police say Best was found "unresponsive" near the garage door exit of an apartment building in Washington, D.C.'s Navy Yard neighborhood at 5:25 a.m. on Nov. 1, and was pronounced dead by medical personnel who responded to the scene.
The city's Office of the Chief Medical Examiner on Thursday said Best died from "multiple blunt force injuries" and it ruled his death a suicide. It would not release further information.
In announcing his death, HHS Secretary Alex Azar said the 49-year-old former CVSHealth and Pfizer Pharmaceuticals executive agreed to work at HHS "out of a desire to serve the American people by making health care more affordable."
"He brought his deep expertise and passion to this task with great humility and collegiality," Azar's statement said. "All of us who served with Dan at HHS and in the administration mourn his passing and extend our thoughts and prayers to his wife Lisa and the entire Best family at this difficult time."
Best is survived by his wife, Lisa, and three children.

The bill would invoke the president’s authorities under the International Emergency Economic Powers Act, a law which allows the White House to intervene in commerce during a national emergency. US entities that continue to do business with ByteDance would be subject to penalties under the proposed legislation.
TikTok spokesperson Brooke Oberwetter was critical of the bill, saying it “takes a piecemeal approach to national security and a piecemeal approach to broad industry issues like data security, privacy, and online harms.”
“We hope that [Hawley] will focus his energies on efforts to address those issues holistically, rather than pretending that banning a single service would solve any of the problems he’s concerned about or make Americans any safer,” the spokesperson added.
While Hawley told reporters on Capitol Hill that his legislation “specifically goes after TikTok” and “bans it,” the language of the bill appears to contain no measure to prohibit the app from American devices. It is unclear how such a ban would be enforced.
In addition to barring transactions with ByteDance and other entities linked with TikTok, the new bill also calls for a report to be compiled by the US intelligence community on the alleged “national security threat posed by TikTok.” The report would focus on whether the Chinese government can use the app to access US data, especially for “intelligence or military purposes,” and outline any “ongoing efforts” by Beijing to monitor or manipulate American citizens online.
Hawley has led a group of lawmakers, most of them Republicans, who are highly critical of TikTok and its Chinese parent firm, with the senator recently calling the app “China’s backdoor into Americans’ lives.” He spearheaded the effort to ban TikTok on some government devices late last year, which received bipartisan support, but has argued the measure did not go far enough.

White House comments on potential NATO-Russia clash
There is “absolutely no indication” that Moscow is planning to strike the US-led military bloc, Washington says
NATO countries are not in danger of being attacked by Russia, US National Security Council spokesman John Kirby said on Wednesday.
Speaking at a regular press briefing, Kirby was asked whether the US-led military alliance has enough troops to repel a possible Russian strike on its eastern flank in case of a major escalation in the Ukraine conflict.
“All I can tell you is we’ve seen absolutely no indication that [Russian President Vladimir] Putin has designs on striking NATO territory,”the spokesman said, while stressing that US President Joe Biden is taking Washington’s Article 5 commitments “seriously.” He was referring to the principle that an attack on one member is an attack on the entire alliance.
Kirby noted that Washington has deployed an additional 20,000 US service personnel in Europe, bringing their total number to 100,000. He added that NATO is “confident that we have the capability, the energy, the talent, the manpower, the resources to meet our Article 5 commitments.”
Russia has consistently described the US military presence on NATO’s eastern flank as a threat. In late October, Kremlin Press Secretary Dmitry Peskov warned that “the closer American troops are to our borders, the greater danger we are in.”
Kirby also said the US is paying attention to the red lines drawn by Moscow in Ukraine, adding: “We don’t just take anything for granted when they say it. And we’re not dismissive.”
In the autumn, Russia’s Foreign Ministry warned Washington that deliveries of long-range weapons to Kiev would cross a “red line” and make America “a direct party to the conflict.” On Tuesday, Dmitry Polyansky, Moscow’s First Deputy Permanent Representative to the UN, suggested that while the West has disregarded some of Moscow’s warnings, “it is possible that the most red [lines] have not been crossed yet”.
However, on Wednesday, Russia’s Foreign Ministry said that all “talk about red lines has become a thing of the past,” pointing to the ongoing “hybrid war” waged by the West against Russia in Ukraine.
“The United States has unequivocally declared that it seeks to inflict a strategic defeat on Russia. It's impossible to ignore reality,” the ministry added.

Vermont Supreme Court Upholds Measure That Allows Non-Citizens To Vote
In 2021, Democrats in Vermont granted non-citizens the right to vote in municipal elections. Republicans filed lawsuits and the legal battle went all the way to the state’s highest court.
Now the Vermont Supreme Court has upheld the measure.
The Associated Press reported:
Vermont Supreme Court rules in favor of noncitizen voting
Noncitizen residents may continue to vote in municipal elections in Vermont’s capital city of Montpelier, the state Supreme Court ruled Friday, stating that such voting in local elections doesn’t violate the state constitution.
In its decision on an appeal the higher court upheld a lower court ruling, dismissing the claim.
“The statute allowing noncitizens to vote in local Montpelier elections does not violate Chapter II, § 42 because that constitutional provision does not apply to local elections,” the Supreme Court wrote.
In 2021, the Democrat-controlled Vermont Legislature approved two separate bills to change the municipal charters of Montpelier and Winooski, the most diverse community in the state, to allow legal residents who are not U.S. citizens to vote in local elections. Republican Gov. Phil Scott vetoed the measures, but the Legislature overrode his vetoes.
The Republican National Committee filed lawsuits against the two Vermont cities asking judges to declare noncitizen voting unconstitutional and lost those challenges. Federal law prohibits noncitizens from voting in federal elections, including races for president, vice president, Senate or House of Representatives.
Montpelier and Winooski are among more than a dozen communities in a handful of states — including New York City, and a number of towns in Maryland — that allow noncitizens to vote in local elections. Seven states, including Ohio, have measures banning noncitizen voting in local elections, according to the group Americans for Citizen Voting.

DANVILLE, Pa. — Three monkeys that had been lost along the interstate in Montour County are all accounted for, all of them now dead.
A public health risk assessment was conducted by several organizations including the CDC.
They confirm the escaped monkeys were humanely euthanized.
The monkeys escaped after a truck carrying about 100 of them collided with a dump truck Friday afternoon along Route 54 just off Interstate 80 near Danville.
"We're rural Pennsylvania. We hear about like cows getting out and you know pigs and trucks like that wrecking, but Monkeys it's not a common everyday thing," said Jessica Wright, Danville.
According to a CDC spokesperson, the monkeys were en route to a CDC-approved quarantine facility after landing Friday morning in New York. They are originally from Mauritius, a country in eastern Africa.
The crash happened around 3:30 p.m. Friday afternoon, that's when the three monkeys got free in the Danville area.
Police say the driver who was transporting the monkeys was charged in the crash.
Originally, state police told us four monkeys were on the loose.
As of Saturday afternoon, some people were still worried.
"Little monkeys, we got bears, we have coyotes, we have deer, you know all the time. A little 3-pound monkey doesn't scare me, but why are they so concerned about it is what concerns me," said Howie Lerch, Valley Township.
Friday night Newswatch 16 spoke with Michelle Fallon of Danville, who saw the entire accident. She jumped into action; helping both drivers and the loads they were carrying.
"I walk up back on the hill and this guy tells me, 'Oh, he's hauling cats. I said, 'oh.' So I go over to look in the crate and there's this green cloth over it. So I peel it back, I stick my finger in there and go 'kitty, kitty.' It pops its head up and it's a monkey," Fallon said.
Fallon was contacted Saturday by the CDC and was told to monitor herself for any cold-like symptoms.
She shared the letter from the CDC with Newswatch 16; it reads in part that, "the surviving monkeys will be quarantined and will be monitored for infectious diseases for at least 31 days before their release."

The European Union should move its economy to a wartime footing amid the Ukraine conflict, Manfred Weber, the president of the European People’s Party (EPP), suggested on Thursday. He said that the move is required to ramp up the bloc's own defense potential, and that of Kiev.
Speaking to the newspapers of the Funke media group, Weber called for the reorganization of the EU arms industry. The MEP believes that the bloc needs “a kind of war economy… in order to be able to guarantee stability and security”.
“The European states are currently not in a position to provide the necessary armaments quickly enough, neither for our own defense, nor for Ukraine,” he lamented. To remedy the situation, Weber argued, the EU should “break new ground,” pointing out that complex allocation structures are not working the way they are supposed to.
The MEP pointed out that Germany had established a €100 billion ($109 billion) special defense fund amid the Ukraine conflict, but “hardly anything happens because the bureaucracy is paralyzing.” “A year after the start of the war, the armaments companies have hardly received any orders,” he added.

On Wednesday, commenting on the Western tank shipments, Kremlin Press Secretary Dmitry Peskov reiterated that this armor “would burn as [did] the rest of it.”

Poland allegedly gave Ukraine several MiG-29 planesunder the guise of spare parts
In spring 2022,Warsaw secretly delivered several of its MiG-29 fighter jets to Ukraine, despite the Polish government officially denying any such deals, a local paper has claimed, citing sources.
According to Dziennik Gazeta Prawna (DGP), the planes were sent over using a “combined” method, apparently meaning that they were delivered in a disassembled state and declared as spare parts.
“The fuselage and the wings are also spare parts,” DGP wrote Wednesday, citing sources within the Polish government.
Back in March, in the first months of Russia’s military operation in Ukraine, Washington rejected a plan to transfer Polish MiG-29 jets to Ukraine, stating the move was “too escalatory” and risked directly involving the US or a NATO ally in the conflict, potentially triggering a direct confrontation with Russia.
In April, however, the Pentagon stated that unnamed US allies had helped bolster Ukraine’s fleet of fighter jets by donating unspecified “spare parts” which were supposedly used to restore many of Kiev’s damaged planes.
The latest article by DGP now seems to suggest that that ally could have been Poland, which in late April was also revealed to have provided Ukraine with $7 billion worth of military aid, including half of its tanks, dozens of howitzers, Grad MRLS, and missiles for MiG-29 and Su-27 fighter jets, among other munitions.
In recent weeks the US seems to have abandoned some of its prior concerns about supplying heavy weapons to Ukraine, and now plans to deliver a number of its M1 Abrams tanks to Kiev, while Germany, Poland and Finland intend to send dozens of their Leopard 2 tanks.
Kremlin spokesperson Dmitry Peskov has responded to these planned deliveries as evidence that the West is becoming more directly involved in the conflict, despite European and American politicians claiming otherwise. That’s as Russia has repeatedly urged the West to stop “pumping” Ukraine with weaponry, arguing it would only prolong the conflict and lead to more bloodshed.

The most advanced mind-controlled devices being tested in humans rely on tiny wires inserted into the brain. Now researchers have paved the way for a less invasive option. They’ve used ultrasound imaging to predict a monkey’s intended eye or hand movements—information that could generate commands for a robotic arm or computer cursor. If the approach can be improved, it may offer people who are paralyzed a new means of controlling prostheses without equipment that penetrates the brain.
“This study will put [ultrasound] on the map as a brain-machine interface technique,” says Stanford University neuroscientist Krishna Shenoy, who was not involved in the new work. “Adding this to the toolkit is spectacular.”
Doctors have long used sound waves with frequencies beyond the range of human hearing to create images of our innards. A device called a transducer sends ultrasonic pings into the body, which bounce back to indicate the boundaries between different tissues and fluids.
About a decade ago, researchers found a way to adapt ultrasound for brain imaging. The approach, known as functional ultrasound, uses a broad, flat plane of sound instead of a narrow beam to capture a large area more quickly than with traditional ultrasound. Like functional magnetic resonance imaging (fMRI), functional ultrasound measures changes in blood flow that indicate when neurons are active and expending energy. But it creates images with much finer resolution than fMRI and doesn’t require participants to lie in a massive scanner.
The technique still requires removing a small piece of skull, but unlike implanted electrodes that read neurons’ electrical activity directly, it doesn’t involve opening the brain’s protective membrane, notes neuroscientist Richard Andersen of the California Institute of Technology (Caltech), a co-author of the new study. Functional ultrasound can read from regions deep in the brain without penetrating the tissue.
Still, gauging neural activity from a distance means sacrificing some speed and precision, says Andersen’s co-author, Caltech biochemical engineer Mikhail Shapiro. Compared with electrodes’ readings, functional ultrasound provides “a less direct signal,” he says, so “there was a question of how much information [ultrasound images] really contain.” The images could reveal neural activity as the brain prepared for a movement. But was there enough detail in that signal for a computer to decode the intended move?
To find out, the researchers slotted small ultrasound transducers, roughly the size and shape of a domino, into the skulls of two rhesus macaque monkeys. The device—attached by a wire to a computer—aimed sound waves down into a region of the brain called the posterior parietal cortex, which is involved in planning movements.

To catch a glimpse of evolution, Pim Stemmer doesn’t have to set sail for the Galapagos. He needn’t trudge through the Costa Rican rain forest or blast into Antarctic sea ice. For a front-row view of evolution in action, Stemmer simply walks over to a lab bench at Maxygen, a biotech startup in Redwood City, Calif., where he oversees research and development. At Maxygen, researchers are tapping evolutionary principles to “breed” new proteins, successively fine-tuning specific traits-say, heat tolerance or the ability to stick to a cancer cell-by tinkering with the underlying genetic code and selecting only the best of the new genes for the next round of improvement. In farm fields and kennels, exploiting evolution is old hat. Farmers favor a corn crop that best survives summer, for instance, and with decades of patience, breeders can produce dogs as specialized as poodles and Pomeranians. But in the lab, evolving better proteins is a recent feat-and a faster one. By focusing their efforts at the genetic level, one molecule at a time, researchers can coax a protein toward perfection in just a matter of weeks. In the process, companies like Maxygen hope to crank out safer medications, more potent cleansers and healthier crops. Just a few years out of the gate, these young firms are already bringing new protein products to market, forging alliances with such giants as Dow Chemical, DuPont and Novartis and launching successful IPOs (see table, “Evolving Industry”)-all evidence that “directed evolution” is really happening, not just in the test tube, but also in the marketplace.
Evolutionary Alternative
Directed evolution is an alternative to “rational protein design, ” a technique that became popular in the 1980s. In rational protein design, researchers try to craft a new molecule-perhaps an antibody or an enzyme-by first studying an existing protein’s structure and then modifying it via targeted mutations to the gene that encodes it. But that sort of painstaking methodology can prove to be difficult. Not only must researchers determine the sequence of amino acids-the 20 building blocks that make up all proteins-but they must also understand the complicated pattern of folding that the chain of amino acids undergoes to become a functional, three-dimensional molecule. Even after bringing sophisticated computer tools to bear on the problem, researchers say, it’s hard to unravel the workings of a protein folded up like origami, much less create a new one that behaves the way you want it to. “Rational design has failed miserably at helping us make useful proteins,” says Caltech’s Frances Arnold, who sits on Maxygen’s scientific advisory board.

Frustrated with the limitations of rational design, a growing group of researchers are borrowing from nature’s tool kit instead, mimicking the basic processes of evolution-the generation of genetic diversity and the selection of desirable traits-to improve proteins, even without understanding their complicated structures. And turning to nature can pay off quickly. A team led by MIT chemical engineer Dane Wittrup, for example, evolved an antibody fragment to bind 10,000 times more tightly to its target in just four rounds of directed evolution. Researchers like Wittrup hope better-binding antibodies might someday be used to fight cancer: Attach a cell-killing agent to an antibody that binds specifically and tightly to molecules found only on cancer cells, the logic goes, and you can wipe out the cancer without damaging healthy tissues. And it’s not just antibodies but a wide range of potentially useful proteins-from industrial enzymes to protein pharmaceuticals-that stand to benefit. “With just a few evolutionary cycles, we can dramatically improve molecules that have frustrated companies for years,” says Stemmer.
Do the Shuffle
Stemmer invented his version of directed evolution, called “molecular breeding,” seven years ago, while working at Affymax, in Palo Alto, Calif. The brainchild of entrepreneur Alejandro Zaffaroni, Affymax was the first company to focus exclusively on discovering new drugs through combinatorial chemistry-a shotgun approach in which huge libraries of unique molecules are randomly generated, and then the useful ones are fished out through clever screening. Extending the combinatorial idea into the realm of protein development, Stemmer struck upon the idea of shuffling DNA. The basic concept: Start with a few different versions of the gene for a protein you’d like to improve, cut them up and mix the pieces together to generate a diverse pool of new versions of the gene, fish out the ones you like best, and start all over again. In 1997, Zaffaroni spun off Maxygen to take molecular breeding to market.
Larger companies, sniffing the commercial potential of evolution in a test tube, began hooking up with Maxygen. One of the first was the Danish firm Novo Nordisk, the largest industrial enzyme maker in the world, which had already been dabbling in directed evolution on its own. Reporting last year in the journal Nature Biotechnology, scientists from the two companies showed how directed evolution might churn out a new laundry detergent enzyme. The researchers began with the structural gene for Savinase, a stain-eating enzyme developed by Novo Nordisk. Next they collected DNA for the naturally occurring version of the enzyme, subtilisin, from 25 different strains of Bacillus bacteria. With scissors-like enzymes, the team chopped up Savinase and wild subtilisin DNA, shuffling it together to create a new generation of unique “daughter” genes. Then the researchers inserted each gene into a bacterial cell. Finally, they exposed those cells to different temperatures and pHs to see how the resulting proteins held up.
The results were intriguing. While Savinase works best in a limited range of conditions-cool water and a fairly alkaline environment-some of the new proteins produced by the experiment worked four times better, and under acidic conditions. Many of the daughter molecules also performed better than their parents when heated or dunked in organic solvents. For 35 years, Stemmer notes, teams of industry scientists have tried to rationally design an improved subtilisin enzyme. “Within a year, three of our scientists turned out a much better molecule,” he says.

Celebrate Diversity
And who couldn’t use a better molecule? Analysts say the market for directed evolution stretches wide, including medicine and agriculture in addition to the chemicals industry. “This is no fad,” says David Molowa, a biotech analyst at Chase Manhattan in New York. “I think this technology is very real, and it’s already generating new products.” This list today includes a potential new catalyst for penicillin production, developed by Maxygen and now in commercial development at Dutch conglomerate DSM, and a fat-stain-removing enzyme from Novo Nordisk called Lipoprime, already on the market.
Indeed, the $2 billion industrial enzyme market is a logical niche for directed evolution, says Carolyn Fritz, global director of industrial biotechnology for Dow Chemical. Fewer than 30 enzymes generate more than 90 percent of industrial enzyme sales. That’s not for lack of trying on the part of the chemical companies to make new ones. The problem is that most enzymes fizzle under harsh, real-world conditions. Manufacturers could use new enzymes to make everything from paper to ethanol, low-cholesterol cooking oils and blue jeans. “We have a lot of different customers who could use better enzymes,” Fritz says.
To bring those enzymes to life, Dow has partnered with another biotech firm, San Diego-based Diversa, which has a different take on directed evolution from Maxygen. Rather than starting with a few different versions of a gene and shuffling the DNA, Diversa researchers typically begin with one gene and then introduce a multitude of mutations.
This technique generates maximum diversity in the pool of new candidate proteins. Indeed, using a procedure called “gene-site saturation mutagenesis,” Diversa researchers can try each of the 20 possible amino acids in each position along the protein chain-in less than two weeks. “It’s a numbers game,” says Dan Robertson, head of enzyme technology at Diversa, a game the company hopes will yield hardier and more effective proteins.
Diversa researchers stack the odds in their favor by choosing unusual genes as starting points. The company has hired far-flung scientists to collect microbes from extreme locations-the gut of a bug from the Costa Rican jungle, an industrial dump site or the rotting skin of a submerged whale carcass. By harvesting DNA from bacteria on that dead whale, Diversa scientists collect the raw genes for enzymes that naturally break down polymers or fats in nasty environments. “If we need a high-temperature enzyme to work under an alkaline pH, we go looking for places that already have those kinds of conditions,” explains Diversa CEO Jay Short. “We discover enzymes that are optimal in those settings, and then we can use directed evolution to push favored traits even further.”
Short says Diversa hopes to spread its discoveries around, selling improved proteins for everything from animal feed to human medicine. “I think directed-evolution tools are going to be very important for evolving human therapeutics-increasing a drug’s binding affinity or its half-life, or lowering the dosages necessary for it to work,” Short says.
Diversa isn’t the only evolutionary firm sizing up the $300 billion worldwide pharmaceuticals market. After

working on enzymes for the oil industry, Enchira Biotechnology of The Woodlands, Texas, is moving toward crafting antibiotics and drugs that fight cancer, says Peter Policastro, company president. San Diego’s Applied Molecular Evolution is packing an extra punch into existing drugs by changing their protein structures, evolving more powerful medicines. Currently, AME is partnering with Gaithersburg, Md.’s MedImmune to churn out second-generation versions of several MedImmune drugs.
For now, Chase Manhattan’s Molowa says that directed-evolution firms will continue to sell themselves as technology-platform providers, partnering with industry giants that promise dollars and distribution. In addition to its deal with Novo Nordisk, Maxygen has forged collaborations with DuPont subsidiary Pioneer Hi-Bred, AstraZeneca and others-and has “proof-of-principle collaborations” with Abbott, Pfizer and Novartis under way. Diversa has joined forces not only with Dow, but with Novartis and Rhone-Poulenc, among others, as well. Eventually, Molowa adds, look for firms like Maxygen and Diversa to roll out their own products.
White Potato, Red Potato
As in nature, however, evolution in the lab can be a bumpy ride. Some researchers caution that the new technique has its limitations. “You’re not going to turn a potato into a broccoli,” says Arnold at Caltech. “Maybe you can turn a white potato into a red potato. The question is, what’s close, in terms of evolution? We just don’t know yet.”
It’s a question lacking easy answers. At Maxygen, company president Simba Gill says DNA shuffling has worked in 90 percent of experiments-and the firm drops the experiments that fail. Duke University biochemist Homme Hellinga says he would like to see chemists combine aspects of rational protein design and directed evolution, using computers to model new proteins and then testing well they can adapt to different environments.
But even proteins that evolve beautifully in the lab face classic development hurdles, warns Arnold. Yet another obstacle could crop up in an arena that is becoming central to almost all of high technology these days: intellectual property. Both Maxygen and Diversa have staked relatively broad patent claims, which some industry observers say might not survive litigation. And a patent issued to AME in 1998, if broadly interpreted, might force some players in the field to seek licenses from AME in order to continue their projects. AME set up a licensing program this spring and has already successfully defended its patent against one challenge in Japan. Regardless of how the legal landscape shapes up, though, directed evolution seems certain to offer researchers in academia and industry alike a faster route to better proteins for years to come.

The movement focuses on anti-vaccine narratives based on unfounded claims that getting blood from persons who have been immunized against the coronavirus "contaminates" the body.
Some have campaigned for blood banks that draw from "pure" unvaccinated people, and North American medics say they have received requests for transfusions from donors who have not gotten the vaccine.
Vaccine skeptics, dubbed "pure bloods," incite violence against doctors who provide coronavirus vaccines in closed social media forums, amid bogus claims of mass deaths of vaccinated people.
The recent high-profile instance of a New Zealand couple seeking to deny life-saving heart surgery for their infant on the grounds that any blood transfused could have originated from a vaccinated donor heightened the panic.
Their attitude forced a New Zealand court to temporarily take custody of the baby in order to facilitate the treatment, but the story became a cause celebre among vaccine skeptics worldwide.
According to the Safe Blood website, participants must pay an initial joining cost of 50 euros ($54), followed by a yearly fee of 20 euros.
In the United States, when more than 80% of the population has had at least one Covid shot, the Food and Drug Administration states that blood collectors are not required to screen for vaccination status.
Hospitals are also unable to advise patients about the status of donated blood.
