HEK293 Protocols: Cell Culture, Transfection, Protein Production

HEK 293 cells are popular for their ease of growth and transfection (HEK293 Transfection Kit), making them a common cell culture in cancer research. In addition, high transfection efficiency of HEK293 cells produces exogenous proteins or viruses for pharmaceutical and biomedical research purposes. HEK-293 cells are useful for many transfection experiments, particularly the propagation of adenoviral-based and retroviral-based vectors.

Origins of the HEK293 Cell Line

HEK293 is a cell line derived from human embryonic kidney cells grown in tissue culture. They are also known, more informally, as HEK cells. This particular line was initiated by the transformation and culturing of normal HEK cells with sheared adenovirus 5 DNA. The transformation resulted in the incorporation of approximately 4.5 kilobases from the viral genome into human chromosome 19 of the HEK cells. The line was cultured by scientist Alex Van der Eb in the early 1970s at his lab at the University of Leiden, Holland. The transformation was executed by Frank Graham, another scientist Van der Eb’s lab who invented the calcium phosphate method for transfecting cells. The source of the cells was a healthy aborted fetus of unknown parenthood. The name HEK293 is thusly named because it was Frank Graham’s 293rd experiment.

The type of kidney cell that the HEK293 cell line came arose from is unknown and it is difficult to conclusively characterize the cells post-transformation since adenovirus 5 could have significantly disrupted cell morphology and expression. Also, embryonic kidneys are a heterogeneous mix of almost all the types of cells present in the body. In fact, it has been speculated by independent researchers, including Van der Eb himself, that the cells may be neuronal in origin. Although theoretically possible, most cells derived from an embryonic kidney would be endothelial, epithelial or fibroblast cells. Neuronal origin is suspected due to the presence of mRNA and gene products typically found in neurons.

Today, HEK293 cells are frequently used in cell biology and biotechnology, second only to HeLa, the first human cell line. Around establishment of HeLa in 1951, scientists were reluctant to accept and use human cell lines out of concern for an oncogenic agent in them. This concern, along with the known ability of animal cell lines to grow rapidly and yield a high amount of proteins, gave scientists reason to favor animal cell lines over human cell lines when producing recombinant proteins. However, advances in technology since then have allowed for an increase in human cell line use. One advantage of human cell lines is that they are able to produce proteins most similar to those that humans naturally synthesize. Now there are approved recombinant biotherapeutic products produced from HEK293 and other human cell lines.

HEK293 and its derivatives are used in a wide range of experiments, including signal transduction and protein interaction studies, rapid small-scale protein production, and biopharmaceutical production. HEK293 cells easily grow in suspension serum-free culture, reproduce rapidly, and produce high levels of protein, which explains why they have been widely used to produce research-grade proteins for a number of years.

Family of Marine killed in Afghanistan sues Alec Baldwin for defamation

The family of a Marine killed in Afghanistan during the Biden administration’s chaotic troop withdrawal has sued Alec Baldwin for defamation, claiming the actor falsely accused one of the serviceman’s grieving sisters of being an “insurrectionist.”

Rylee McCollum, 20, of Wyoming, was one of 13 US service members killed in the Aug. 26 bombing outside the Abbey Gate of Hamid Karzai International Airport, where they were processing would-be evacuees to be flown out of Afghanistan following the Taliban takeover.

President Joe Biden, who promised to unite the nation in his inaugural address one year ago, has failed in epic fashion in the eyes of voters, according to a series of year one polls out this week.

In the latest, 60% called the president’s first year unsuccessful. Worse, said Rasmussen Reports, 50% said Biden has been “very unsuccessful,” and a third of Democrats also think he’s failed.

Add to that a new Gallup survey that found Biden’s promise of unity broken in pieces. “Party polarization highest for first-year president,” it headlined, noting an 83-point divide in his approval ratings between Democrats and Republicans. It was 75 points at the end of former President Donald Trump’s first year.

And Morning Consult had his approval rating at the outfit’s lowest, 40%.

Many of the pollsters have pointed out that Trump’s numbers were slightly worse than Biden’s, but they leave out that fewer expected Trump to do well, while most expected Biden, a vice president for eight years and a senator for decades, would succeed with ease.

Crushing Biden’s numbers has not been the job of Republicans but independents who have abandoned him and exasperated Democrats. Morning Consult said on Wednesday, “Biden is entering his second year in office with his worst job approval rating yet, driven by increasingly tepid support among Democrats that comes as his agenda remains stalled on Capitol Hill.”

Rasmussen’s year in review poll showed Biden’s approval with independents upside down. Just 30% see his presidency as successful versus 69% who don’t.

And the pollster also looked at the president’s unity pledge and, like Gallup, found it to be a failure. Just 12% said the nation was more united, while 57% said it was more divided.

The timing could not be worse for Biden and the Democrats. Without the public's support, the president has been unsuccessful in wooing two holdout senators to back his agenda, and 2022 midterm election predictors are pointing to a GOP blowout.

Conspiracy projection has split apart the country. The Left has fought efforts to learn the full truth, as they project conspiracies to disguise conspirators.

American politics over the last half decade has become immersed in a series of conspiracy charges leveled by Democrats against their opponents that, in fact, are happening because of them and through them. The consequences of these conspiracies becoming reality and reality revealing itself as conspiracy have been costly to American prestige, honor, and security. As we move away from denouncing realists as conspiracists, and self-pronounced “realists” are revealed as the true conspirators, let’s review a few of the more damaging of these events.

Russians on the Brain

Consider that the Trump election of 2016, the transition, and the first two years of the Trump presidency were undermined by a media-progressive generated hoax of “Russian collusion.”

The “bombshell” and “walls are closing in” mythologies dominated the network news and cable outlets. It took five years to expose them as rank agit-prop.

Robert Mueller and his “dream team” consumed $40 million of Americans’ money and 22 months of our time—to find the nothingburger that most of the country already knew was nothing. Yet the subtext of the 2018 Democratic takeover of the House was the media narrative that Trump, as Hillary Clinton put it, was an “illegitimate” president, due to Russian collusion.

Former Director of National Intelligence James Clapper claimed on national television that Trump was a “Russian asset.” Former CIA head John Brennan assured the nation that the president was “treasonous,” due to his supposed “lies to the American people.”

All sorts of politicians, retired military, and news anchors echoed the charges. The lies and myths of has-been British spy Christopher Steele made him a leftist hero. They were repeated ad nauseam as truth.

FBI grandees like James Comey, Andrew McCabe, Peter Strzok, Lisa Page, Kevin Clinesmith, and others destroyed their careers in their obsessions over Trump. In the process of destroying themselves, they also nearly wrecked the reputation of the FBI. The Pentagon has the lowest popular support in modern memory. The CIA is more feared by millions of Americans than by our enemies.

Steele could not produce any evidence to back up his scurrilous charges. The inspector general of the Department of Justice found little evidence to substantiate any of the charges. James Comey and Robert Mueller under oath both pleaded either memory problems or denied any knowledge about the FBI’s use of Steele and the role his fake dossier played.

Most of the televised accusers, when under oath before Congress, admitted they had no evidence for their flamboyant charges. Representative Adam Schiff (D-Calif.), who claimed the dossier was authentic and that Trump was compromised, has repeatedly been revealed as a liar. In various televised House hearings, he kept getting caught either fabricating, misrepresenting, or warping evidence before his own committee.

No matter. Hillary Clinton and the Left kept pounding “collusion.” The more it was proven false, the more they shouted the lie.

Finally, special counsel John Durham’s investigations, some authentic media investigative reporting, and the preponderance of evidence showed not only that Trump did not collude with the Russians, but that the entire charge was a sick sort of projection on the part of Hillary Clinton and her vassals.

Steele concocted the election-cycle fantasy using a former Clintonite totem in Moscow. Another source was the now indicted Igor Danchenko ( a “primary sub-source”), who was working at the leftist Brookings Institution while feeding Steele.

A cynic might look at this sad chapter and conclude that Hillary Clinton sought to destroy her opponent by paying Christopher Steele to manufacture fantasies fabricated from left-wing and former Clinton associates. Then she used the media, the FBI, the CIA, and the Justice Department to seed the farce while hiding her own role behind three firewalls including the DNC, the Perkins Coie law firm, and Glenn Simpson’s Fusion GPS.

The ultimate irony?

Hillary Clinton did collude with those claiming to have Russian connections to warp an election, and she projected her likely illegal activity onto the target of her attacks. No conspiracist could trump such reality. Will Merrick Garland look at her role in this episode as he conducts his insurrectionist investigations concerning efforts to undermine an election?….
