I don't see why I should care. Though last week my biophysics professor went on a tangent during class that the fact that we can't measure entropy suggests that we haven't really understood the underlying physics.

Not directly, I think. You can calculate it by measuring other stuff, eg you can measure Gibbs free energy and you can measure enthalpy and from the difference between the two you get entropy.
