Konosuba isn't specifically parodying or subverting anything in particular, it's just comedy adventure story set in a fantasy world. The isekai aspect of it is played pretty straight aside from the comedic way Kazuma dies, and you don't really need to be familiar to the common tropes of isekai, fantasy games or any of that to find the comedy funny because most of the humor comes from the characters and the silly world it's set in rather than from parody or subverting expectations.
