The Japanese name for the Scepter, the energy meter, and Island in the game is "Kidoairakku", which is a mashup of the 4 powers Peach has in the game, so "Joyragegloomcalm". Peachette as a concept is slightly different then what transpired in Super Mario 64 DS. In the latter, Yoshi could pick up the hat of Mario, Luigi or Wario, which in turn would turn him completely into that character. That character would show no traces of being Yoshi except for the voice, which seems to imply that it's not much more then a glorified costume. With Peachette, it's still evident that it still retains some element of being Toadette, thanks to the name, and the design elements that carry over that are more commonly associated with either Toads themselves or Toadette (like the shoes), yet her voice also completely changes along with the power up despite still being Toadette. This isn't another character turning into another character in appearance and abilities only, this is still Toadette, but turned into a different form of Toadette that happens to look and sound like Peach.
Obviously this element was added into the New NSMBU port so that they could create a half-hearted excuse for people to use a Peach-like character without having to contradict the story of the game, as Peach is kidnapped and should not be able to be playable. So it's just a "psuedo-Peach" that's a passable replacement to dodge the fact that Peach is kidnapped. Since Toadette is another popular female Mario character, she was the best character used as a vessel for this idea so that people can still play "a Peach" while Peach herself is still held by Bowser and you don't deal with the awkward scenario of just having 2 completely identical Peaches present (like what they did in Luigi in Super Mario galaxy, even if they still kind of explained it in that game).
So with the explanation for this being obvious, why is it still a big deal and rife with theory crafting? Well, a common mystery in the Mario universe that many always wondered was about Peach's relationship to the Toad's (and lesser extent Goombas) that was always shrugged off because "well it's Mario, who cares? lmao" so many didn't ever bother to dig deep into it. Goombas are said to be traitors of the Mushroom Kingdom and signed up with Bowser. This explains their Mushroom-like appearance and why they contrast so much with the rest of the turtle-like Koopas, as they too used to be natural inhabitants of the Mushroom Kingdom. Toads, obviously, are also very mushroom-like, and are seemingly the main inhabaitants of the Mushroom Kingdom. Many of Peach's servants are Toads, and her personal assistant is an elderly Toad name Toadsworth. Peach is completely surrounded by Toads, not humans like herself. Within her castle, within various depictions of towns and locations appearing in various Mario games all shows this fact that humans seem to be pretty rare in the Mushroom Kingdom (or at least within close proximity of Peach's Castle). In fact, the only human-like characters you really ever see her deal with in the main games is Mario and Luigi. Why is such a human-looking character that shows no traces of being a mushroom or shaped like one is the one ruling over a nation full of mushroom like people? This Peachette form, Toadette literally powering up which makes her look like a very human form resembling Peach, could give multiple explanations to why Peach looks human while the rest of the toads stay Toads throughout their entire lives. This is why everyone now all of a sudden cares about it and others worry about the Game Theory-tier theorizing that will soon follow because of it.
It doesn't help that every other human Mario character has plausible deniability when it comes to them looking like humans that can be explained away a bit easier then Peach can.

Mario and Luigi could be argued that they didn't grow up in the Mushroom Kingdom. Construction Site from Donkey Kong and the location of Wrecking Crew can be speculated to all take place in the same City-like area. If so, this area is likely the Big City, the first major level of Donkey Kong for the Gameboy which features a New York-like city. Most would think so in the beginning, but with the release of Odyssey and it's structure of "Kingdom's" with Pauline as the head seem to imply that it may be outside of the Mushroom Kingdom. If so, then Mario & Luigi originally being Toads may not be true if they originated from outside the Mushroom Kingdom. (However, a major strike against this theory is that Yoshi's Island shows that at the end of the game as a baby they were dropped from the Stork to parents that lived in Mushroom shaped houses. So maybe they did grow up in the Mushroom Kingdom, but could have grew up in the City-like area for most of their lives.)
Wario and Waluigi also have deniability. They have pointed ears, which is far different then any other human Mario character we've seen since. It's very possible that they are a different race of humans altogether, having more goblin-like or elf like genes then Mario and Luigi. It can be argued that whatever race of humans they are could have no relations to Toads, as we don't see it anywhere being reproduced outside of Wario and Waluigi.
Princess Daisy is not a native of the Mushroom Kingdom, she is the ruler of Sarasaland. The locations that appear in Sarasaland also take heavy influence from our real world, and I don't believe they show any signs of Toads there, so naturally Daisy is assumed to be fully human.

So what is the deal? I have some theories, hopefully none not too out there.
>1. Queen Toadstool. One theory can be similar to determination of a "Queen" in some insect creatures. Throughout maybe a variety of factors, there is a chance that any born female Toad will not be toad-looking at all, but have genetic coding for a more human-like form, and in their culture this determines the ruler of the nation and inherit the royal lineage. To go even deeper into insectoids, they could be responsible for all reproduction that still births out Toads while still looking like a full human. Remember how Nintendo tried to claim how Toad's are genderless? So how do they reproduce? Most came to the conclusion after that revelation was that they just did what Mushrooms did and shedd spores, but it could be that they don't reproduce at all. If they don't, then it could still explain why they still exist as a species while there's another "Queen" that is in charge of reproducing.
>2. Evolution. Toads will eventually naturally evolve into humans, and Peach is the showcase of the final stages of this evolution, and Peachette skipped this whole process for Toadette. But this explanation brings issues. Why is Peach completely evolved into a human while the rest of the Toads have not? You would think it's not only Peach that have gone through this full evolution.
>3. The Crown. Peach may still be a toad, similar to Toadette, but the power of the crown she wears changes her appearance. Again, this could be a natural thing for the rulers of the Kingdom to do, or it could be that Peach is trying to connect more with the likes of Mario, Luigi, and Daisy who are for this theory under the assumption to be more fully human. This is the most literal explanation of the mystery surrounding Peach, as it mirrors exactly what Toadette goes through in the NSMBU Switch port: she puts on a crown and turns into a Peach-like. Peach could have gone through the exact same thing. Therefore, Peach's natural form could still be a Toad that we've never seen (This also seems to imply that the humanoid "Peach" look that both Peach and Peachette showcase is not unique, which is quite distressing to think deeply about)
>4. Conquerors. Similar to #4, Peach is not a Toad, and may not even be an natural inhabitant of the Mushroom Kingdom, but her family rules the Kingdom because of a past conquering of the land. What lead me to include this theory here was seeing old pictures of the King and Queen of the Mushroom Kingdom. While very rare for them to be referenced in Mario these days, so much so that one could think they were retconned, during the early years of the franchise they did exist, mostly the King and especially during Super Mario Bros and Super Mario Bros. 3 era. They too are often depicted as humans with very little mushroom characteristics and generally look different from Toads. Maybe the entire family are not naturally Toads, and somehow gained rulership of the Kingdom still many years ago and now rule through family inheritance. This would give adequate explanation to why Peach looks absolutely not like any of the Toads.
>5. The Stork. Similar to #4, Peach is not a toad and may not have even been born from Toads. As I referenced earlier for Mario and Luigi, Yoshi's Island DS also showed that Peach was delivered by the Stork, and it could be possible that she has no relation to the toads because of this.
